Synaptic Vesicle Dynamics sans Dynamin [PDF]
The neuron-specific guanosine triphosphatase dynamin 1 has been hypothesized to be critically required for pinching off synaptic vesicles during endocytosis. In a recent publication in Science, Ferguson et al. describe a series of experiments demonstrating an unexpectedly selective requirement of dynamin 1 in synaptic vesicle endocytosis only during ...

Background Lymphotoxin β receptor (LTβR) is a member of tumor necrosis factor receptor (TNFR) superfamily which regulates the immune response. At the cellular level, upon ligand binding, the receptor activates the pro-inflammatory NF-κB and AP-1 pathways.

Calcineurin Selectively Docks with the Dynamin Ixb Splice Variant to Regulate Activity-dependent Bulk Endocytosis [PDF]
Depolarization of nerve terminals stimulates rapid dephosphorylation of two isoforms of dynamin I (dynI), mediated by the calcium-dependent phosphatase calcineurin (CaN).

Delivery of Pleckstrin‐Homology Domains Suppresses PI3K/Akt Signaling and Breast Cancer Metastasis
Current therapies curb tumor growth but not metastasis. Obscurin, a giant metastasis suppressor lost in breast cancer, restrains PI3K/Akt signaling but is impractical to restore. We deploy a mini‐obscurin, comprising the obscurin‐PH‐domain, which sequesters PI3K‐p85, potently suppressing invasion and metastasis.

Dynamin and receptor‐mediated endocytosis [PDF]
The internalization of receptor‐bound ligands involves concentration of cell surface receptors in specialized areas of the plasma membrane and subsequent formation of clathrin‐coated vesicles. The complex process of invagination, constriction and budding of clathrin‐coated vesicles employs the coordinated actions of several proteins.

The dynamin inhibitor, dynasore, prevents zoledronate-induced viability loss in human gingival fibroblasts by partially blocking zoledronate uptake and inhibiting endosomal acidification [PDF]
Objective For treatment of medication-related osteonecrosis of the jaw, one proposed approach is the use of a topical agent to block entry of these medications in oral soft tissues.

Ubiquitination in Control of EGF Receptor Endocytosis [PDF]
Growth factors and growth factor receptors comprise a signaling network regulating cell differentiation and growth. Tightly regulation of this network is important to avoid excessive signaling and cell growth, which may result in cancer.

Coacervate‐Mediated Lysosome‐Targeting Antibody Delivery for Protein Degradation
Targeting coacervates to the lysosome in the cytoplasm is a challenge. Here, we convert a tetrapeptide into lysosome‐targeting, membrane‐translocating coacervates. The lysosome‐sorting peptide coacervates facilitate coacervate‐mediated delivery of antibody antigen complexes and a PROTAC compound to the lysosome for the targeted degradation of cancer ...

Membrane curvature controls dynamin polymerization [PDF]
The generation of membrane curvature in intracellular traffic involves many proteins that can curve lipid bilayers. Among these, dynamin-like proteins were shown to deform membranes into tubules, and thus far are the only proteins known to mechanically drive membrane fission.

Toxoplasma and Plasmodium protein kinases: roles in invasion and host cell remodelling [PDF]
Some apicomplexan parasites have evolved distinct protein kinase families to modulate host cell structure and function. Toxoplasma gondii rhoptry protein kinases and pseudokinases are involved in virulence and modulation of host cell signalling.
