Protein pyrophosphorylation by inositol pyrophosphates — detection, function, and regulation
Protein pyrophosphorylation is an unusual signaling mechanism that was discovered two decades ago. It can be driven by inositol pyrophosphate messengers and influences various cellular processes. Herein, we summarize the research progress and challenges of this field, covering pathways found to be regulated by this posttranslational modification as ...

Balitora dehouensis, a new species of cave-dwelling hillstream loach (Teleostei, Cypriniformes, Balitoridae) from the upper Red River, southwestern China [PDF]
A new species of cave-dwelling hillstream loach (Balitoridae, Cypriniformes), Balitora dehouensis sp. nov. is described from a cave in Wenshan Zhuang and Miao Autonomous Prefecture, Yunnan Province, China. Balitora dehouensis sp. nov.

Function‐driven design of a surrogate interleukin‐2 receptor ligand
Interleukin (IL)‐2 signaling can be achieved and precisely fine‐tuned through the affinity, distance, and orientation of the heterodimeric receptors with their ligands. We designed a biased IL‐2 surrogate ligand that selectively promotes effector T and natural killer cell activation and differentiation. Interleukin (IL) receptors play a pivotal role in

Early growth response‐1 induction by fibroblast growth factor‐1 via increase of mitogen‐activated protein kinase and inhibition of protein kinase B in hippocampal neurons [PDF]
Background and purpose: The transcription factor early growth response‐1 (Egr‐1) and the acidic fibroblast growth factor (FGF‐1) are involved in many regulatory processes, including hippocampus‐associated learning and memory. However, the intracellular signalling mechanisms regulating Egr‐1 in hippocampal cells are not entirely understood.Experimental

Time after time – circadian clocks through the lens of oscillator theory
Oscillator theory bridges physics and circadian biology. Damped oscillators require external drivers, while limit cycles emerge from delayed feedback and nonlinearities. Coupling enables tissue‐level coherence, and entrainment aligns internal clocks with environmental cues.

The newfound relationship between extrachromosomal DNAs and excised signal circles
Extrachromosomal DNAs (ecDNAs) contribute to the progression of many human cancers. In addition, circular DNA by‐products of V(D)J recombination, excised signal circles (ESCs), have roles in cancer progression but have largely been overlooked. In this Review, we explore the roles of ecDNAs and ESCs in cancer development, and highlight why these ...

Cryptochrome and PAS/LOV proteins play intricate roles in circadian clocks where they act as both sensors and mediators of protein–protein interactions. Their ubiquitous presence in signaling networks has positioned them as targets for small‐molecule therapeutics. This review provides a structural introduction to these protein families.

This study reveals how the mitochondrial protein Slm35 is regulated in Saccharomyces cerevisiae. The authors identify stress‐responsive DNA elements and two upstream open reading frames (uORFs) in the 5′ untranslated region of SLM35. One uORF restricts translation, and its mutation increases Slm35 protein levels and mitophagy.

Essential regulatory elements for NHE3 gene transcription in renal proximal tubule cells
The objectives of the present study were to identify the cis-elements of the promoter absolutely required for the efficient rat NHE3 gene transcription and to locate positive and negative regulatory elements in the 5’-flanking sequence (5’FS),
