EF-hand proteins are homologous; they have all evolved from a common precursor. They contain from two to twelve copies of the EF-hand domain. The EF-hand is about thirty amino acids long and consists of an α-helix (E), loop, and second α-helix (F). Usually a Ca2+ ion is bound in the loop under physiological conditions; however, 30% of all known EF ...

Tryptophan Scanning Mutagenesis of EF-Hand Motifs
2019Ca2+ regulation in living systems occurs via specific structural alterations, subtle or drastic, in the Ca2+-binding domains of sensor proteins. Sensor proteins perform designated nonredundant roles within the dense network of Ca2+-binding proteins. A detailed understanding of the structural changes in calcium sensor proteins due to Ca2+ spikes that ...

The EF-hand family of calcium-modulated proteins
Trends in Neurosciences, 1989The EF-hand homolog proteins bind calcium (Ca2+) with dissociation constants in the micromolar range and are modulated by stimulus-induced increases in cytosolic free Ca2+. We have grouped over 160 different EF-hand homolog proteins into ten subfamilies and ten unique categories.

Engineering new metal specificity in EF-hand peptides
JBIC Journal of Biological Inorganic Chemistry, 2003Peptides (33-34 amino acids long) corresponding to the helix-turn-helix (EF-hand) motif of the calcium binding site I of Paramecium tetraurelia calmodulin have been synthesized. The linear sequence was unable to acquire a native-like conformation and calcium binding.

Mutations of Gln41 and Lys75 with nonpolar residues in the N-terminal domain of calmodulin (N-Cam) revealed the importance of solvation energetics in conformational change of Ca(2+) sensor EF-hand domains. While in general these domains have polar residues at these corresponding positions yet the extent of their conformational response to Ca(2 ...

The EF-Hand, Homologs and Analogs
1991Many proteins bind calcium; they have different structures and employ different coordination geometries to achieve a wide range of affinities and selectivities. Kretsinger (1975) proposed that those within the cytosol are calcium modulated, that they are in the magnesium or apoform in the quiescent cell and in the calcium form in the stimulated cell ...
