A highly chemo- and enantioselective PTC alkylation has been developed that allows rapid access to orthogonally protected (S,S)-isodityrosine. Utility of this material in the construction of isodityrosine-containing cyclic peptides is demonstrated by synthesis of the natural product renieramide.

Enantioselective synthesis of levomilnacipran

A novel approach for the asymmetric synthesis of the active (1S,2R)-enantiomer of the antidepressant milnacipran is reported. The two stereogenic centers borne by the cyclopropane ring were sequentially installed starting from phenylacetic acid.

Enantioselective metabolism of cumene

1. The enantioselective metabolism of cumene (isopropylbenzene) was studied in intact rabbits. 2. Of the total 2-phenyl-1-propanol formed metabolically, 90.3% was shown by h.p.l.c. to be (R)-(+)-2-phenyl-1-propanol. The corresponding values for (S)-(+)-2-phenylpropanoic acid and (R)-(-)-2-hydroxy-2-phenylpropanoic acid were 99.0 and 81.0%, respectively.
