Purification and Some Properties of Endo-polygalacturonase from Tomato Pericarp
トマト果肉より5%食塩(pH6.0)でPGを抽出し, 硫安分画, SP-Sephadexカラムクロマトグラフィー, Sephadex G-100のゲル濾過により約48.6倍に精製した.精製酵素はディスク電気泳動および免疫電気泳動的に均一であり, ゲル濾過法により測定した分子量は, 52, 000であった.本酵素には3.39%の糖が含有されており, ポリガラクチュロン酸を基質とした場合, Km値は0.267%であった.また食塩で活性が増大し, 0.15M食塩で約3倍の活性増大がみられた.本酵素は基質の粘度を急激に低下させるEndo-PGである.
