Nano‐Enabled Toxicity of Pharmaceutical Excipients

open access: yesWIREs Nanomedicine and Nanobiotechnology, Volume 18, Issue 3, May/June 2026.
Excipients regarded as safe in bulk form may become toxic once incorporated into nanocarriers. This review raises awareness of nano‐enabled toxicity and highlights analytical tools and safer alternative excipients to support risk‐mitigated nanomedicine development.

Endolysosomal trapping of therapeutics and endosomal escape strategies

Drug Discovery Today
Internalizing therapeutic molecules or genes into cells and safely delivering them to the target tissue where they can perform the intended tasks is one of the key characteristics of the smart gene/drug delivery vector. Despite much research in this field, endosomal escape continues to be a significant obstacle to the development of effective gene/drug

Endosomal escape pathways for delivery of biologicals

Journal of Controlled Release, 2011
Despite continuous improvements in delivery systems, the development of methods for efficient and specific delivery of targeted therapeutic agents still remains an issue in biological treatments such as protein and gene therapy. The endocytic pathway is the major uptake mechanism of cells and any biological agents, such as DNA, siRNA and proteins ...

Unravelling the endosomal escape of pH-responsive nanoparticles using the split luciferase endosomal escape quantification assay

Biomaterials Science
Our work demonstrates the adaptation of a highly versatile split-luciferase endosomal escape quantification (SLEEQ) assay to better understand the endosomal escape efficiency of pH-responsive nanoparticles by tuning the HiBiT linker chemistry.
