Loss of primary cilia promotes EphA2‐mediated endothelial‐to‐mesenchymal transition in the ovarian tumor microenvironment

open access: yesMolecular Oncology, EarlyView.
Loss of primary cilia in endothelial cells promotes EndMT and vascular abnormalities in the ovarian tumor microenvironment through EphA2 activation. Using human samples, in vitro models, and endothelial‐specific Kif3a‐knockout mice, we show that primary cilia loss drives the acquisition of cancer‐associated fibroblast‐like phenotypes, thereby ...

A vascular endothelial growth factor–loaded chitosan-hyaluronic acid hydrogel scaffold enhances the therapeutic effect of adipose-derived stem cells in the context of stroke

open access: yesNeural Regeneration Research
Adipose-derived stem cell, one type of mesenchymal stem cells, is a promising approach in treating ischemia-reperfusion injury caused by occlusion of the middle cerebral artery.

Efficient Differentiation of Human Pluripotent Stem Cells to Endothelial Cells [PDF]

open access: yesCurrent Protocols in Human Genetics, 2018
AbstractEndothelial cells (ECs) line the interior surface of blood and lymphatic vessels, and play a key role in a variety of physiological or pathological processes such as thrombosis, inflammation, or vascular wall remodeling. Human‐induced pluripotent stem cell (iPSCs)‐derived ECs provide a new opportunity for vascular regeneration and serve as a ...

Endoglin mediates the tumor‐ and metastasis‐promoting traits of stromal myofibroblasts in human breast carcinomas

open access: yesMolecular Oncology, EarlyView.
Carcinoma‐associated fibroblasts (CAFs) in tumors influence cancer progression. We identified endoglin (ENG) as a key factor in TGF‐β signaling in myofibroblastic CAFs (myCAFs), linked to poor breast cancer outcomes. Inhibiting ENG on myCAFs suppressed the TGF‐β‐Smad2/3 pathway, reducing primary tumor growth and metastasis.

Host-microbe interactions at the blood-brain barrier through the lens of induced pluripotent stem cell-derived brain-like endothelial cells

open access: yesmBio
Microbe-induced meningoencephalitis/meningitis is a life-threatening infection of the central nervous system (CNS) that occurs when pathogens are able to cross the blood-brain barrier (BBB) and gain access to the CNS.

Understanding and measuring mechanical signals in the tumor stroma

open access: yesFEBS Open Bio, EarlyView.
This review discusses cancer‐associated fibroblast subtypes and their functions, particularly in relation to extracellular matrix production, as well as the development of 3D models to study tumor stroma mechanics in vitro. Several quantitative techniques to measure tissue mechanical properties are also described, to emphasize the diagnostic and ...

Bone Morphogenetic Protein 9 Regulates Early Lymphatic-Specified Endothelial Cell Expansion during Mouse Embryonic Stem Cell Differentiation

open access: yesStem Cell Reports, 2019
Summary: Exogenous cues involved in the regulation of the initial steps of lymphatic endothelial development remain largely unknown. We have used an in vitro model based on the co-culture of vascular precursors derived from mouse embryonic stem cell (ESC)

Retinoic Acid Promotes Endothelial Cell Cycle Early G1 State to Enable Human Hemogenic Endothelial Cell Specification

open access: yesCell Reports, 2020
Summary: Development of blood-forming (hemogenic) endothelial cells that give rise to hematopoietic stem and progenitor cells (HSPCs) is critical during embryogenesis to generate the embryonic and postnatal hematopoietic system.

Mesenchymal Stem Cells Can Be Differentiated Into Endothelial Cells In Vitro [PDF]

open access: yesSTEM CELLS, 2004
Human bone marrow-derived mesenchymal stem cells (MSCs) have the potential to differentiate into mesenchymal tissues like osteocytes, chondrocytes, and adipocytes in vivo and in vitro. The aim of this study was to investigate the in vitro differentiation of MSCs into cells of the endothelial lineage.

Bioengineering facets of the tumor microenvironment in 3D tumor models: insights into cellular, biophysical and biochemical interactions

open access: yesFEBS Open Bio, EarlyView.
The tumor microenvironment is a dynamic, multifaceted complex system of interdependent cellular, biochemical, and biophysical components. Three‐dimensional in vitro models of the tumor microenvironment enable a better understanding of these interactions and their impact on cancer progression and therapeutic resistance.
