Enzymic hydrolysis of steroid phosphates

Abstract Steroid phosphates were used as substrates for alkaline and acid phospliatases from different sources to determine the structural specificity of the compounds that undergo enzymic hydrolysis. The steroid 21-, 3β- and 3α-phosphates were hydrolyzed by the acid and alkaline phosphatases tested.

An improved enzymic hydrolysis of collagen

Abstract A peptidase mixture was prepared from hog-kidney microsomes which contained aminopeptidase, prolidase, prolinase, carboxypeptidase-P, and X-prolyl-aminopeptidase activity. The sequential addition of collagenase, papain, and the peptidase mixture to collagen samples resulted in the hydrolysis of 90% of the peptide bonds.

Hydrolysis of peptide esters by different enzymes

The combined use in peptide synthesis of the Fmoc‐group with methyl, benzyl or p‐nitro benzyl esters is not practical because of the elimination of the Fmoc‐group under basic conditions and by catalytic hydrogenation. Nevertheless the solution synthesis of peptides requires those combinations in some cases.

Abstract The hydrolysis ofN:N-dimethylcarbamoyl fluoride by rabbit tissues and by various vertebrate plasmata has been studied. The hydrolysis of this compound by rabbit liver and kidney homogenates involves an enzyme distinct from that hydrolysing di iso propoxyphosphoryl fluoride.

Ceramide glucoside (1-O-glucosido-2-N-acyl-sphingosine) was hydrolysed to ceramide (N-acyl-sphingosine) and glucose by beta-glucosidase from ox brain. The reaction was stimulated by the non-ionic detergent, Triton X-100, or by the anionic detergents, cholate or taurocholate.

Ceramide lactoside [1-O-(galactosido-4-beta-glucosido)-2-N-acyl-sphingosine] was hydrolysed to ceramide glucoside and galactose by beta-galactosidase of rat brain. The reaction was not reversible, required cholate or taurocholate, had optimum pH5.0 and K(m) 2.2x10(-5)m.

The hydrolysis of ochratoxin a by some proteolytic enzymes

Ochratoxin A, a metabolite produced by a strain of Aspergillus ochraceus and causing enteritis, renal necrosis, an increase of glycogen in the liver when given to rats, as well as inhibiting the phosphorylase enzyme in vitro, is found to be hydrolyzed in vivo to an iso-coumarin derivative (ochratoxin a).

The hydrolysis of xylan and xylooligosaccharides by ruminal enzymes

Abstract Xylan is hydrolyzed to a mixture of xylooligosaccharides by enzymes in the rumen of the bovine. A random hydrolysis of the xylosidic bonds of xylan, xylotetraose, xylotriose, and xylobiose is effected. Several bacterial cultures obtained from the rumen contents were shown to produce xylan-hydrolyzing enzymes.
