Spreading of EGF Receptor Activity into EGF-free Regions and Molecular Therapies of Cancer [PDF]

open access: yesarXiv, 2008
The primary activation of the epidermal growth factor receptor (EGFR) has become a prominent target for molecular therapies against several forms of cancer. But despite considerable progress during the last years, many of its aspects remain poorly understood.

ShcD adaptor protein drives invasion of triple negative breast cancer cells by aberrant activation of EGFR signaling

open access: yesMolecular Oncology, EarlyView.
We identified adaptor protein ShcD as upregulated in triple‐negative breast cancer and found its expression to be correlated with reduced patient survival and increased invasion in cell models. Using a proteomic screen, we identified novel ShcD binding partners involved in EGFR signaling pathways.

A case of multiple primary lung adenocarcinoma with a CD74‐NRG1 fusion protein and HER2 mutation benefit from combined target therapy

open access: yesThoracic Cancer, 2022
Neuregulin 1 (NRG1) gene fusion is a rare oncogenic driver gene in multiple tumor types, leading to the activation of the epidermal growth factor receptor (ErbB)‐mediated pathway.

A Radiogenomics Pipeline for Lung Nodules Segmentation and Prediction of EGFR Mutation Status from CT Scans [PDF]

open access: yesarXiv, 2022
Lung cancer is a leading cause of death worldwide. Early-stage detection of lung cancer is essential for a more favorable prognosis. Radiogenomics is an emerging discipline that combines medical imaging and genomics features for modeling patient outcomes non-invasively.

Structure of the Epidermal Growth Factor Receptor Kinase Domain Alone and in Complex with a 4-Anilinoquinazoline Inhibitor*

open access: yesJournal of Biological Chemistry, 2002
The crystal structure of the kinase domain from the epidermal growth factor receptor (EGFRK) including forty amino acids from the carboxyl-terminal tail has been determined to 2.6-Å resolution, both with and without an EGFRK-specific inhibitor currently ...

Targeting the AKT/mTOR pathway attenuates the metastatic potential of colorectal carcinoma circulating tumor cells in a murine xenotransplantation model

open access: yesMolecular Oncology, EarlyView.
Dual targeting of AKT and mTOR using MK2206 and RAD001 reduces tumor burden in an intracardiac colon cancer circulating tumor cell xenotransplantation model. Analysis of AKT isoform‐specific knockdowns in CTC‐MCC‐41 reveals differentially regulated proteins and phospho‐proteins by liquid chromatography coupled mass spectrometry. Circulating tumor cells

Exploiting memory effects to detect the boundaries of biochemical subnetworks [PDF]

open access: yesarXiv, 2023
Partial measurements of biochemical reaction networks are ubiquitous and limit our ability to reconstruct the topology of the reaction network and the strength of the interactions amongst both the observed and the unobserved molecular species. Here, we show how we can utilize noisy time series of such partially observed networks to determine which ...

Time, the final frontier

open access: yesMolecular Oncology, EarlyView.
This article advocates integrating temporal dynamics into cancer research. Rather than relying on static snapshots, researchers should increasingly consider adopting dynamic methods—such as live imaging, temporal omics, and liquid biopsies—to track how tumors evolve over time.

When Two are Better than One: Modeling the Mechanisms of Antibody Mixtures [PDF]

open access: yes, 2019
It is difficult to predict how antibodies will behave when mixed together, even after each has been independently characterized. Here, we present a statistical mechanical model for the activity of antibody mixtures that accounts for whether pairs of antibodies bind to distinct or overlapping epitopes.
