The Effects of Anticonvulsant Drugs on NMDA-EPSP, AMPA-EPSP, and GABA-IPSP in the Rat Hippocampus

Brain Research Bulletin, 1997
The effects of phenobarbital, phenytoin, and valproic acid on pharmacologically isolated NMDA-EPSP, AMPA-EPSP, and GABA-IPSPs were examined in rat hippocampal slices. Phenobarbital (0.05 mg/ml) had no effect on the NMDA-EPSP, but decreased the slope of the AMPA-EPSP by 13.4% and facilitated the GABA-IPSP slope by 77.12%.

The Catalytic Mechanism of EPSP Synthase Revisited

Biochemistry, 1999
Recent analysis of EPSP synthase by solid-state NMR has led to the postulation of a new enzyme reaction pathway and raised once again the question of an intermediate species covalently bound to the enzyme [Studelska, D., McDowell, L., Espe, M., Klug, C., and Schaefer, J. (1997) Biochemistry 36, 15555-15560].

Transition State Analysis of Enolpyruvylshikimate 3-Phosphate (EPSP) Synthase (AroA)-Catalyzed EPSP Hydrolysis

Journal of the American Chemical Society, 2012
Proton transfer to carbon atoms is a significant catalytic challenge because of the large intrinsic energetic barrier and the frequently unfavorable thermodynamics. The main catalytic challenge for enolpyruvylshikimate 3-phosphate synthase (EPSP synthase, AroA) is protonating the methylene carbon atom of phosphoenolpyruvate, or EPSP, in the reverse ...

Reevaluating Glyphosate as a Transition-State Inhibitor of EPSP Synthase: Identification of an EPSP Synthase.cntdot.EPSP.cntdot.Glyphosate Ternary Complex

Biochemistry, 1995
Numerous studies have confirmed that glyphosate forms a tight ternary complex with EPSP synthase and shikimate 3-phosphate. It has been proposed [Anton, D., Hedstrom, L., Fish, S., & Abeles, R. (1983) Biochemistry 22, 5903-5908; Steinrücken, H. C., & Amrhein, N. (1984) Eur. J. Biochem.

Inhibition of 5-enolpyruvoylshikimate 3-phosphate (EPSP) synthase by analogs of the tetrahedral intermediate and of EPSP

Journal of the American Chemical Society, 1992
A number of analogues of the intermediate and product of the enzyme EPSP synthase are described. Inert analogues that show high affinity for the enzyme from Petunia hybrida are produced by replacing the labile ketal phosphate moiety of this intermediate with a phosphonate or by stabilizing the ketal phosphate against ionization through introduction of ...

Construction and assessment of reaction models of Class I EPSP synthase. Part II: investigation of the EPSP ketal

Journal of Biomolecular Structure and Dynamics, 2013
Although the proposed mechanisms are reasonable, there are still many questions about the 5-enolpyruvyl shikimate-3-phosphate (EPSP) synthase mechanism that are difficult to answer by experimental means alone. EPSP synthase is a key enzyme in the shikimic acid pathway, which is found only in plants and some micro-organisms and is also molecular target ...

Reduced corticomotoneuronal excitatory postsynaptic potentials (EPSPs) with normal Ia afferent EPSPs in amyotrophic lateral sclerosis

Neurology, 1996
We studied excitatory postsynaptic potentials (EPSPs) arising in single spinal motoneurons (composite EPSPs) induced by Ia afferent and magnetic cortical stimulation in 28 normal subjects ranging in age from 24 to 84 years and 28 patients with amyotrophic lateral sclerosis (ALS) aged 34 to 82 years. The subjects voluntarily recruited single motor units

Further results with the ‘uniquantal EPSP’ hypothesis

Hearing Research, 1997
The hypothesis of Geisler (Brain Res. 212 (1981) 198-201), in which the different spontaneous-rate classes of primary auditory neurons were accounted for by the different sizes of uniquantal EPSPs relative to the gap between resting membrane and threshold potentials, was represented with an expanded model which included relative refractory effects. The

Metabotropic glutamate receptor dependent EPSP and EPSP-spike potentiation in area CA1 of the submerged rat hippocampal slice

Journal of Neurophysiology, 1996
1. We reexamined the important areas of conflict in (1S,3R)-1-aminocyclopentane-1,3-dicarboxylic acid [(1S,3R)-ACPD]-induced potentiation of the field excitatory postsynaptic potential (EPSP) and, for the first time, investigated the role of mGluRs in EPSP-spike (E-S) coupling. 2. (1S,3R)-ACPD (10 microM) bath applied for 20 min consistently induced a
