Covalent drug discovery: Progress against key targets, emerging strategies and lessons learnt

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 22, Page 6574-6606, November 2026.
Abstract Covalent drug discovery is currently experiencing a boom in industrial and academic interest. To date, at least 75 covalent drugs have received regulatory approval, targeting both traditional target classes and more challenging proteins for which other approaches failed. In many cases, unique aspects of covalent targeting are essential for the

Tackling FGFR3-driven bladder cancer with a promising synergistic FGFR/HDAC targeted therapy

open access: yesnpj Precision Oncology, 2023
Bladder cancer (BC) is one of the most prevalent malignancies worldwide and FGFR3 alterations are particularly common in BC. Despite approval of erdafitinib, durable responses for FGFR inhibitors are still uncommon and most patients relapse to metastatic

Next‐Generation Sequencing‐Based Molecular Profiling and Management of Advanced Solid Tumors: A Real‐World Experience in a Community‐Based Academic Cancer Center

open access: yesCancer Medicine, Volume 15, Issue 10, October 2026.
ABSTRACT Background Advances in molecular and cell biology have led to a paradigm shift in cancer management. We implemented next‐generation sequencing (NGS) at our community‐based academic cancer center over a decade ago. We sought to optimize the use of genomics and understand its impact in identifying actionable genomic alterations to determine ...

Patient‐Derived Organoids in Functional Precision Oncology: Tumor Modeling, Drug‐Response Prediction, and Clinical Translation

open access: yesCell Biochemistry and Function, Volume 44, Issue 10, October 2026.
ABSTRACT Patient‐derived organoids (PDOs) have emerged as an effective experimental platform for bridging the long‐standing gap between traditional in vitro cancer models and the biological complexity of actual tumors. These three‐dimensional cultures (3D), derived directly from patient tissues, preserve essential architectural, genomic, and phenotypic

Oncogenic Characterization and Pharmacologic Sensitivity of Activating Fibroblast Growth Factor Receptor (FGFR) Genetic Alterations to the Selective FGFR Inhibitor Erdafitinib

open access: yes, 2017
Fibroblast growth factor receptor (FGFR) genetic alterations are frequently observed in cancer, suggesting that FGFR inhibition may be a promising therapy in patients harboring these lesions. Identification of predictive and pharmacodynamic biomarkers to

Neuro‐associated SEMA3C+ tumour cells promote gastric cancer peritoneal metastasis via EBF2–FGF7–FGFR2‐mediated tumour–endothelial crosstalk

open access: yesClinical and Translational Medicine, Volume 16, Issue 10, October 2026.
Single‐nucleus sequencing identified a semaphorin 3C (SEMA3C)‐high stem‐like tumour subpopulation that drives gastric cancer peritoneal metastasis through early B‐cell factor 2 (EBF2)‐mediated FGF7 expression and enhanced FGF7–FGFR2 signalling with endothelial cells. Targeting the FGF–FGFR pathway may provide a promising therapeutic strategy.
