Flap Endonuclease Activity of Gene 6 Exonuclease of Bacteriophage T7

open access: yes, 2019
Background: Flap endonucleases remove 5-single-stranded DNA termini. Results: T7 gene 6 exonuclease is a flap endonuclease that cleaves 5-single-stranded termini one nucleotide into the duplex.

Some Tuco‐Tucos Hide More Than Others: An Integrative Approach Reveals Cryptic Diversity in Ctenomys (Rodentia: Ctenomyidae) From Central Argentina

open access: yesIntegrative Zoology, EarlyView.
Here, we adopt an integrative framework (molecular phylogenetics and geometric morphometrics) to reassess patterns of diversity and differentiation across this region. We analyzed cytochrome b sequences and quantified cranial shape variation. Our analyses indicate the presence of three species groups (mendocinus, talarum, and tucumanus) and two ...

Covalent drug discovery: Progress against key targets, emerging strategies and lessons learnt

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Covalent drug discovery is currently experiencing a boom in industrial and academic interest. To date, at least 75 covalent drugs have received regulatory approval, targeting both traditional target classes and more challenging proteins for which other approaches failed. In many cases, unique aspects of covalent targeting are essential for the

Kinetics and thermodynamics of DNA polymerases with exonuclease proofreading

open access: yes, 2016
Kinetic theory and thermodynamics are applied to DNA polymerases with exonuclease activity, taking into account the dependence of the rates on the previously incorporated nucleotide.

Decreased Expression by a Chemically Modified siRNA to PTBP1 Seriously Affects Warburg Effect in Colorectal Cancer Cells

open access: yesCancer Science, EarlyView.
Chemically modified siR‐PTBP1 disrupts the Warburg effect by reprogramming pyruvate kinase splicing from PKM2 to PKM1. This metabolic shift activates the TCA cycle, increases ATP production, and excessive ROS generation, and ultimately induces apoptotic cell death in colorectal cancer cells.

Genetic Engineering of Tumor‐Infiltrating Lymphocytes (TIL) via a T‐Editor Platform to Enhance Anti‐Tumor Activity

open access: yesCancer Science, EarlyView.
A rapid and efficient CRISPR‐mediated gene editing platform for TIL engineering identified FAM84B as a novel potential target to enhance antitumor activity and pioneered the use of CBE to generate FAM84B loss‐of‐function TIL with enhanced antitumor activity.

A phase II clinical trial of toripalimab in advanced solid tumors with polymerase epsilon/polymerase delta (POLE/POLD1) mutation

open access: yesSignal Transduction and Targeted Therapy
Patients carrying mutations in polymerase epsilon/polymerase delta have shown positive responses to immune checkpoint inhibitors. Yet, prospective trials exploring the efficacy in those with polymerase epsilon/polymerase delta mutations are still lacking.

The epigenetic landscape of skeletal muscle in response to exercise and aging

open access: yesThe FEBS Journal, EarlyView.
Epigenetic mechanisms regulate gene expression in response to exercise and aging, thereby supporting skeletal muscle plasticity. Acute exercise induces chromatin remodeling through histone modifications and DNA methylation, promoting the expression of exercise‐responsive genes.

A novel TREX1 inhibitor, VB-85680, upregulates cellular interferon responses.

open access: yesPLoS ONE
Activation of the cGAS-STING pathway plays a key role in the innate immune response to cancer through Type-1 Interferon (IFN) production and T cell priming.

Cas9/sgRNAs protect lambda DNA from exonuclease digestion.

open access: yes, 2019
(A) Diagram of lambda genomic DNA shows the relative positions of three Cas9/sgRNA complexes (Lambda F2, Lambda R6, and Lambda R4) and the 12 kb fragment protected by Lambda F2 and Lambda R6 or the 36 kb fragment protected by Lambda F2 and Lambda R4 ...
