Sensitization of immunoresistant prostate carcinoma cell lines to Fas/Fas ligand-mediated killing by cytotoxic lymphocytes: Independence of de novo protein synthesis

We recently reported that drug-resistant prostate tumor cells (DU145, PC-3) are resistant to Fas-mediated killing by cytotoxic lymphocytes, and that this resistance can be overcome by treatment with subtoxic concentrations of chemotherapeutic drugs. Fas belongs to the tumor necrosis factor (TNF) family of receptors.

[Evaluation of restenosis by serum levels of soluble Fas, Fas ligand and nuclear matrix protein before and after coronary intervention].

Serum levels of soluble Fas, soluble Fas ligand, and nuclear matrix protein (NMP) were measured in 38 patients with ischemic heart disease before and after coronary angiography or coronary intervention. Serum levels of soluble Fas, soluble Fas ligand and NMP were determined by enzyme-linked immunosorbent assay.

Effective immunomodulation to induce tolerance to tissue/organ allografts is attained by infusion of donor lymphocytes endowed with killing capacity through ectopic expression of a short-lived Fas-ligand (FasL) protein. The same approach has proven effective in improving hematopoietic stem and progenitor cell engraftment.

Cytotoxic T lymphocytes (CTL) can kill tumor cells and cells infected with intracellular pathogens. Their two major killing mechanisms are the degranulation pathway and the Fas-FasL pathway. The degranulation process consists of the release of cytolytic perforin and granzyme molecules that are stored within lysosomal granules.

The reconstructive plasmid pCEP4x was transfected into HepG2 cells by electroportion method.
