Insecticide Resistance and Fitness Cost
The intensive use of chemicals through decades has been selecting resistant popula‐ tions of several insect species to distinct classes of insecticides, like neurotoxics, in‐ sect growth regulators, and toxins derived from bacteria. Insecticide resistance is nowadays a huge challenge for control programs of pests of rural practices and principally to ...

Time‐resolved X‐ray solution scattering captures how proteins change shape in real time under near‐native conditions. This article presents a practical workflow for light‐triggered TR‐XSS experiments, from data collection to structural refinement. Using a calcium‐transporting membrane protein as an example, the approach can be broadly applied to study ...

Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

A Cost of Sexual Attractiveness to High-Fitness Females
Adaptive mate choice by females is an important component of sexual selection in many species. The evolutionary consequences of male mate preferences, however, have received relatively little study, especially in the context of sexual conflict, where males often harm their mates.

Rifampicin Resistance: Fitness Costs and the Significance of Compensatory Evolution [PDF]
Seventy years after the introduction of antibiotic chemotherapy to treat tuberculosis, problems caused by drug-resistance in Mycobacterium tuberculosis have become greater than ever. The discovery and development of novel drugs and drug combination therapies will be critical to managing these problematic infections.

This study investigated a novel WST‐8‐based assay for evaluating d‐Amino acid oxidase (DAO) inhibitors. We confirmed its effectiveness using known inhibitors and found that uremic toxins possess relatively weak inhibitory activity compared to existing drugs.

Fitness Cost of Chromosomal Drug Resistance-Conferring Mutations [PDF]
ABSTRACTTo study the cost of chromosomal drug resistance mutations to bacteria, we investigated the fitness cost of mutations that confer resistance to different classes of antibiotics affecting bacterial protein synthesis (aminocyclitols, 2-deoxystreptamines, macrolides).

Optimizing photoactivation of PA‐mCherry for optical pooled CRISPR screens
Photoactivatable PA‐mCherry finds widespread use to optically tag individual cells. However, confocal 405 nm UV laser‐scanning (normal scan) is much less efficient than widefield UV illumination, limiting the use of PA‐mCherry on confocal instruments. We remedy this limitation by reporting that rapid and repeated confocal scanning with a low‐intensity,

Bacterial regulatory small RNAs (sRNAs) have shown promise for gene knock‐down studies and metabolic engineering. However, some mRNAs might be difficult to target due to poor binding by the Hfq chaperone, individual synthetic sRNAs can have off‐target ...

Balancing the fit and logistics costs of market segmentations
Segments are typically formed to serve distinct groups of consumers with differentiated marketing mixes, that better fit their specific needs and wants. However, buyers in a segment are not necessarily geographically closely located. Serving a geographically dispersed segment with one marketing mix can increase the logistics costs in the form of high ...
