Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.

Progressive gradient walk for neural network fitness landscape analysis
Understanding the properties of neural network error landscapes is an important problem faced by the neural network research community. A few attempts have been made in the past to gather insight about neural network error landscapes using fitness ...

Inferring genetic interactions from comparative fitness data
Darwinian fitness is a central concept in evolutionary biology. In practice, however, it is hardly possible to measure fitness for all genotypes in a natural population.

Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

Developmental feedbacks and the emergence of individuality
Behavioural individuality is a hallmark of animal life, with major consequences for fitness, ecology, and evolution. One of the most widely invoked explanations for this variation is that feedback loops between an animal's behaviour and its state (e.g ...

Interpreting the effects of DNA polymerase variants at the structural level
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Local Fitness Landscape Exploration Based Genetic Algorithms [PDF]
Genetic algorithms (GAs) have been used to evolve optimal/sub-optimal solutions of many problems. When using GAs for evolving solutions, often fitness evaluation is the most computationally expensive, and this discourages researchers from applying GAs ...

Applications of High-Throughput Sequencing for In Vitro Selection and Characterization of Aptamers
Aptamers are identified through an iterative process of evolutionary selection starting from a random pool containing billions of sequences. Simultaneously to the amplification of high-affinity candidates, the diversity in the pool is exponentially ...

Dormant cancer cells can hide in distant organs for years, evading treatment and the immune system. This review highlights how signals from the surrounding tissue and immune environment keep these cells inactive or trigger their reawakening. Understanding these mechanisms may help develop therapies to eliminate or control dormant cells and prevent ...
