Design and analysis strategies for robust microbiome ageing research

open access: yesFEBS Letters, EarlyView.
The gut microbiome changes with age and associates with age‐related morbidity and mortality, establishing it as a potential biomarker and intervention target for ageing. Realising this potential requires methodological rigour, yet distinguishing biological signals from methodological artefacts remains challenging across cohorts. This review provides an

« Faire connaissance avec la fragilité des choses »

open access: yesRevue d'anthropologie des connaissances
This text is a reflection on material fragilities, grounded in the discussion of two case studies, which look at the reclamation of red mud from bauxite mining in Gardanne and the management of sealed biological samples used in the production of DNA ...

Reconstructing enzyme evolution by protein engineering

open access: yesFEBS Letters, EarlyView.
Natural enzyme evolution can be retraced by protein engineering methods such as directed evolution, rational design, and ancestral sequence reconstruction. These approaches reveal how enzymes emerged from ligand‐binding scaffolds, developed varying substrate preferences, formed oligomeric complexes, adapted to environmental changes, and evolved novel ...

Seismic Fragility Curves of RC Buildings Subjected to Aging

open access: yesGeoHazards
A large number of existing reinforced concrete (RC) buildings have surpassed their anticipated service life and show signs of degradation due to aging; this degradation is a function of the construction practices adopted in the past as well as ...

Reflexivity and fragility

open access: yesEuropean Journal for Philosophy of Science, 2022
Reflexivity is, roughly, when studying or theorising about a target itself influences that target. Fragility is, roughly, when causal or other relations are hard to predict, holding only intermittently or fleetingly. Which is more important, methodologically? By going systematically through cases that do and do not feature each of them, I conclude that

TRAIL‐PEG‐Apt‐PLGA nanosystem as an aptamer‐targeted drug delivery system potential for triple‐negative breast cancer therapy using in vivo mouse model

open access: yesMolecular Oncology, EarlyView.
Aptamers are used both therapeutically and as targeting agents in cancer treatment. We developed an aptamer‐targeted PLGA–TRAIL nanosystem that exhibited superior therapeutic efficacy in NOD/SCID breast cancer models. This nanosystem represents a novel biotechnological drug candidate for suppressing resistance development in breast cancer.

Pre‐analytical optimization of cell‐free DNA and extracellular vesicle‐derived DNA for mutation detection in liquid biopsies

open access: yesMolecular Oncology, EarlyView.
Pre‐analytical handling critically determines liquid biopsy performance. This study defines practical best‐practice conditions for cell‐free DNA (cfDNA) and extracellular vesicle–derived DNA (evDNA), showing how processing time, storage conditions, tube type, and plasma input volume affect DNA integrity and mutation detection.

Fragility Analysis of Masonry Structures Subjected to Random Sequential Ground Motions

open access: yesBuildings
In this study, a fragility analysis of masonry structures is conducted using a random sequential ground motion model. Initially, the relationship between a mainshock and its aftershocks is clarified based on the physical mechanism of “source-path-local ...

IMPDH inhibition enhances cytarabine efficacy in SAMHD1‐expressing leukaemia cells via guanine nucleotide depletion

open access: yesMolecular Oncology, EarlyView.
Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.

"Measuring Macroprudential Risk: Financial Fragility Indexes" [PDF]

open access: yes
With the Great Recession and the regulatory reform that followed, the search for reliable means to capture systemic risk and to detect macrofinancial problems has become a central concern.
