Labeling Deoxyadenosine for the Preparation of Functional Conjugated Oligonucleotides

Bioconjugate Chemistry, 2013
Herein we present a versatile synthetic method for the 8-thioalkylation of (deoxy)adenosine with a short carbon linker having on the other side a variety of molecules (psoralen, acridine) and functional groups (alkyne). After conventional protections, the modified adenosine can be phosphytylated and inserted into an oligonucleotide without affecting ...

Preparation of Antisense Oligonucleotides to Inhibit miRNA Function

Cold Spring Harbor Protocols, 2018
This protocol is used to design antisense oligonucleotides (ASOs) for specific inhibition of miRNA function in cultured cells. The incorporation of 2′-O-methyl-modifications in ASOs enhances their potency and resistance to degradation, whereas 3′-terminal cholesterol-conjugation facilitates delivery of ASOs into cells.

Functional Genomics: High-Density Oligonucleotide Arrays

2002
Publisher Summary As the Human Genome Project and related efforts identify and determine the DNA sequences of human genes, it is important that highly reliable and efficient mechanisms be found to assess individual genetic variation. Three methods for obtaining genome-wide mRNA expression data–oligonucleotide “chips,”’ serial analysis of gene ...

Antagomirzymes: Oligonucleotide Enzymes That Specifically Silence MicroRNA Function

Angewandte Chemie, 2009

Use of Antisense Oligonucleotides in Functional Genomics and Target Validation

Oligonucleotides, 2004
With the completion of sequencing of the human genome, a great deal of interest has been shifted toward functional genomics-based research for identification of novel drug targets for treatment of various diseases. The major challenge facing the pharmaceutical industry is to identify disease-causing genes and elucidate additional roles for genes of ...

Electrochemical Probing of DNA Based on Oligonucleotide-Functionalized Polypyrrole

Biomacromolecules, 2001
We have developed a new type of electrochemical biosensor based on oligonucleotide-functionalized polypyrrole. At first, we analyzed the experimental conditions necessary for building such modified electrodes that show a high electroactivity in aqueous media.

Efficient Functionalization of Oligonucleotides by New Achiral Nonnucleosidic Monomers

Organic Letters, 2014
A novel synthetic strategy has been designed for preparation of achiral nonnucleosidic phosphoramidite monomers for automated solid-phase oligonucleotide synthesis. It is based on O-DMTr-protected 4-(2-hydroxyethyl)-morpholine-2,3-dione as the key compound and a family of building blocks obtained by its ring-opening by primary aliphatic amines.

Versatile Functionalization of Nanoelectrodes by Oligonucleotides via Pyrrole Electrochemistry

ChemPhysChem, 2010
AbstractSurface modification at the nanometer scale is a challenge for the future of molecular electronics. In particular, the precise anchoring and electrical addressing of biological scaffolds such as complex DNA nanonetworks is of importance for generating bio‐directed assemblies of nano‐objects for nanocircuit purposes.

Conjugatable and Bioreduction Cleavable Linker for the 5′-Functionalization of Oligonucleotides

The Journal of Organic Chemistry, 2017
An efficient conjugatable and bioreduction cleavable linker was designed and synthesized for the 5'-terminal ends of oligonucleotides. A phosphoramidite reagent bearing this linker was successfully applied to solid phase synthesis and incorporated at the 5'-terminal ends of oligonucleotides.
