Transgenic overexpression of furin increases epileptic susceptibility [PDF]

open access: yesCell Death & Disease, 2018
AbstractThe proprotein convertase Furin plays crucial roles in the pathology of many diseases. However, the specific role of furin in epilepsy remains unclear. In our study, furin protein was increased in the temporal neocortex of epileptic patients and in the hippocampus and cortex of epileptic mice.

Furin Egress from the TGN is Regulated by Membrane‐Associated RING‐CH Finger (MARCHF) Proteins and Ubiquitin‐Specific Protease 32 (USP32) via Nondegradable K33‐Polyubiquitination

open access: yesAdvanced Science
Furin primarily localizes to the trans‐Golgi network (TGN), where it cleaves and activates a broad range of immature proproteins that play critical roles in cellular homeostasis, disease progression, and infection.

Furin cleavage sites naturally occur in coronaviruses

open access: yesStem Cell Research, 2021
The spike protein is a focused target of COVID-19, a pandemic caused by SARS-CoV-2. A 12-nt insertion at S1/S2 in the spike coding sequence yields a furin cleavage site, which raised controversy views on origin of the virus. Here we analyzed the phylogenetic relationships of coronavirus spike proteins and mapped furin recognition motif on the tree ...

IgG‐Bridging–Seeded Synergistic Aggregation of SARS‐CoV‐2 Spikes Underlies Potent Neutralization by a Low‐Affinity Antibody

open access: yesAdvanced Science, EarlyView.
Low‐affinity antibodies are frequently disregarded in discovery pipelines. This work reports P5‐1C8, a Class 1 SARS‐CoV‐2 antibody with weak trimer binding (KD‐to‐IC50 > 3700‐fold) yet potent neutralization of Omicron JN.1. Structural, biophysical, functional, and coarse‐grained simulations collectively demonstrate that transient inter‐spike IgG ...

Furin cleavage of SARS-CoV-2 Spike promotes but is not essential for infection and cell-cell fusion.

open access: yesPLoS Pathogens, 2021
Severe Acute Respiratory Syndrome coronavirus 2 (SARS-CoV-2) infects cells by binding to the host cell receptor ACE2 and undergoing virus-host membrane fusion.

Membralin Assembles a MAN1B1–VCP Complex to Target Foreign Glycoproteins from the Endoplasmic Reticulum to Lysosomes for Degradation

open access: yesAdvanced Science, EarlyView.
This study identifies Membralin as an ER‐phagy receptor that recruits MAN1B1 and VCP to form a selective ERLAD complex. By sensing dense N‐glycan clusters on viral fusion glycoproteins, this ubiquitin‐independent pathway directs SARS‐CoV‐2 spike, Ebola GP, influenza HA, and HIV‐1 Env to lysosomal degradation, thereby limiting viral infectivity ...

The role of furin in papillomavirus infection [PDF]

open access: yesFuture Microbiology, 2009
Papillomaviruses represent a medically important virus family. Infection with a high-risk human papillomavirus type is a prerequisite for cervical carcinoma development. Infection by low-risk types may result in the generation of benign skin warts. It was recently found that infectious entry of these viruses is dependent upon a specific proteolytic ...

Cyclic Peptidic Furin Inhibitors Developed by Combinatorial Chemistry

open access: yesACS Medicinal Chemistry Letters, 2023
Furin is a human serine protease responsible for activating numerous physiologically relevant cell substrates and is also involved in the development of various pathological conditions, including inflammatory diseases, cancers, and viral and bacterial infections.

Ontogeny of RSPO1, FOXL2, and RUNX1 during ovarian differentiation in the marsupial tammar wallaby

open access: yesDevelopmental Dynamics, EarlyView.
Abstract Background RSPO1 and FOXL2 are female sex‐determining genes involved in the differentiation and organization of the ovary in some eutherian mammals. Mutations or loss of function of these genes are associated with partial to full sex reversal in mice, humans, and goats.

Secretion of Human Furin into Mouse Milk [PDF]

open access: yesJournal of Biological Chemistry, 1997
We have previously described the expression of the human proprotein convertase furin or paired basic amino acid-cleaving enzyme, in mice transgenic for paired basic amino acid-cleaving enzyme and human Protein C (HPC). Here we show 100-fold or higher expression of furin in the mammary gland, compared with endogenous furin.
