Mapping Causal Biology: Mendelian Randomization in the Era of Big Data

open access: yesMed Research, EarlyView.
Mendelian randomization (MR) leverages genetic variants to mitigate confounding biases in causal inference. This review systematically maps MR's methodological evolution, highlights its expanding applications in epidemiology and drug target validation, and outlines future directions for overcoming current biases through dynamic, multi‐omics, and cross ...

Potent Inhibitors of Furin and Furin-like Proprotein Convertases Containing Decarboxylated P1 Arginine Mimetics [PDF]

open access: yesJournal of Medicinal Chemistry, 2009
Furin belongs to the family of proprotein convertases (PCs) and is involved in numerous normal physiological and pathogenic processes, such as viral propagation, bacterial toxin activation, cancer, and metastasis. Furin and related furin-like PCs cleave their substrates at characteristic multibasic consensus sequences, preferentially after an arginine ...

Genome Expansion of ZPB1(a) and ZPC1(a) in Basal Species or Liver Expression of ZPB1a and ZPC1aa in Advanced Species, Two Different Strategies to Ensure Sufficient ZP Synthesis in Teleosts

open access: yesIntegrative Zoology, EarlyView.
We found that teleosts employed two different strategies to ensure sufficient ZP (zona pellucida) synthesis: expansion of ZPB1(a) and ZPC1(a) in basal species or liver expression of ZPB1a and ZPC1aa in advanced species. The N‐terminus of expanded ZPB1(a) or ZPC1(a) and liver‐expressed ZPB1a contains repeated units or low‐complexity regions to form ...

New Treatment Strategy and Future Research Direction for BRAF‐Mutated Cancer

open access: yesCancer Science, EarlyView.
Treatment with BRAF inhibitor plus MEK inhibitor is currently used in BRAF‐mutated various malignancies except colorectal cancer, and treatments with BRAF and/or MEK inhibitors and anti‐EGFR antibody are used in BRAF‐mutated colorectal cancer. Despite recent advances in BRAF‐targeted therapies, their efficacy is still limited.

The R203W substitution drives PACS‐1 syndrome by disrupting intramolecular regulation

open access: yesThe FEBS Journal, EarlyView.
The middle region (MR) of PACS‐1 controls engagement with specific partner proteins. This manuscript presents the structure of the Furin binding region (FBR) and how interactions with partners are regulated through the interplay between a basic patch in the FBR and an acidic cluster in the MR.

Furin Functions as a Nonproteolytic Chaperone for Matrix Metalloproteinase-28: MMP-28 Propeptide Sequence Requirement

open access: yesBiochemistry Research International, 2011
Although MMP-28 is involved in numerous important physiologic and pathologic conditions, the mechanisms of action of this secreted proteinase is not well understood.

Secretion of Human Furin into Mouse Milk [PDF]

open access: yesJournal of Biological Chemistry, 1997
We have previously described the expression of the human proprotein convertase furin or paired basic amino acid-cleaving enzyme, in mice transgenic for paired basic amino acid-cleaving enzyme and human Protein C (HPC). Here we show 100-fold or higher expression of furin in the mammary gland, compared with endogenous furin.

Pan-Cancer Analysis of FURIN as a Potential Prognostic and Immunological Biomarker

open access: yesFrontiers in Molecular Biosciences, 2021
BackgroundFurin is a calcium-dependent protease that processes various precursor proteins through diverse secretory pathways. The deregulation of FURIN correlated with the prognosis of patients in numerous diseases.

Broadening horizons: Pathogenesis and therapeutics of renal ciliopathies

open access: yesJournal of Cell Communication and Signaling, Volume 20, Issue 3, September 2026.
This review elucidates the molecular mechanisms and aberrant signaling pathways in renal ciliopathies, links genetic heterogeneity to clinical phenotypes, and lays a theoretical basis for prenatal diagnosis and novel therapies. Abstract Renal ciliopathies encompass a spectrum of genetic disorders arising from structural or functional impairments of ...
