Mice that are genetically deficient in UDP-galactose: ceramide galactosyltransferase are unable to synthesize galactosylceramide. Consequently, sulfatide, which can be synthesized only by sulfation of galactosylceramide, is also totally absent in affected mouse brain.

Acyl structure regulates galactosylceramide's interfacial interactions
Biochemistry, 1993Galactosylceramides (GalCer) with homogeneous acyl chains containing zero, one, or two cis double bonds have been synthesized and characterized at an argon-aqueous buffer interface using a Langmuir film balance. Both surface pressure and surface potential were measured as a function of molecular area at 24 degrees C. N-Lignoceroylgalactosylsphingosine (

A fluorescent derivative of galactosylceramide [NBD-GalCer: 12-N-methyl-N- (7-nitrobenz-2-oxa-1,3-diazol-4-yl)aminodecanoyl galactosylceramide] was dispersed in sodium dodecylsulfate and used to determine the activity of galactosylceramide-β-galactosidase.

Effect of Galactosylceramide on the Dynamics of Cholesterol-Rich Lipid Membranes
The Journal of Physical Chemistry B, 2011We use atom-scale molecular dynamics simulations to clarify the role of glycosphingolipids in the dynamics of cholesterol-rich lipid rafts. To this end, we consider lipid membranes that contain varying amounts of galactosylceramide (GalCer), sphingomyelin, cholesterol, and phosphatidylcholine.

Glucosylceramide and Galactosylceramide Synthase
2006Most mammalian glycosphingolipid formation begins with the synthesis of cerebrosides, glucosylceramide and galactosylceramide. The synthases catalyzing the formations of glucosylceramide (GlcCer) and galactosylceramide (GalCer) have been characterized at the gene and protein levels.

Clinical Trials with α-Galactosylceramide (KRN7000) in Advanced Cancer
2011For over a century, research has sought ways to boost the immune system of cancer patients in order to eradicate tumors that arose after escaping presumptive immunosurveillance. With increasing knowledge of the immune system, immunotherapeutic strategies to break tolerance to the tumor evolved from largely nonspecific to more specific and increasingly ...

Synthesis of a glucosylated α-S-galactosylceramide as potential immunostimulant
Carbohydrate Research, 2017Synthesis of a glucosylated α-S-galactosylceramide (1), a potential immunostimulant, was achieved starting from D-galactose. Both O- and S-glycosidic linkages were constructed in highly stereoselective way, and the synthetic strategy could be extended to the synthesis of other α-S-GalCer analogues.

Unique sensitivity to α-galactosylceramide of NKT cells in the uterus
Cellular Immunology, 2002It was previously reported that NKT cells, which are mainly present in the liver of mice, are also present in the uterus and that these uterine NKT cells are associated with abortion under overactivated conditions. In this study, we further examined their phenotypic and functional properties. In parallel with the progression of pregnancy, the number of

Distribution of cholesterol and galactosylceramide in rat cerebellar white matter
Biochimica et Biophysica Acta (BBA) - Molecular and Cell Biology of Lipids, 2006White matter and the inner granular layer of rat cerebellum was analysed by imaging time-of-flight secondary-ion mass spectrometry (TOF-SIMS) equipped with a Bi+ ion cluster gun. Samples were prepared by high pressure freezing, freeze-fracturing and freeze drying or by plunge freezing and cryostat sectioning.

Efficient synthesis of galactosylceramide analogues for iNKT cell stimulation
Bioorganic & Medicinal Chemistry Letters, 2010Glycolipids are potential antigens for iNKT cells recognition and demonstrate important roles in both innate and adaptive immunity. However, the difficulties in the preparation of pure configuration defined glycolipids limit the exploration of their different profiles in activating iNKT cells.
