Total Synthesis of (+)- and (-)-Galanthamine
HETEROCYCLES, 2010The stereoselective total synthesis of (+)-galanthamine [(+)-1], an antipode of the natural product, and (-)-galanthamine [(-)-1] starting from D -glucose is described. The cyclohexene unit in (+)-1 was prepared in an optically active form from D -glucose using Ferrier's carbocyclization reaction, and the benzylic quaternary carbon was ...

Facile synthesis of (±)‐, (+)‐, and (‐)‐galanthamine
Journal of Heterocyclic Chemistry, 1995AbstractThe Amarylidacea alkaloid galanthamine (1a) is an acetylcholinesterase inhibitor that has been evaluated as a potential agent for the treatment of Alzheimer's disease. We report a very efficent synthesis of (±)‐galanthamine [(±)‐1a] from readily available isovanillin and tyramine.

Galanthamine in Alzheimer’s Disease
1994Substantial efforts have been made in neurobiology and molecular biology but we are still far from a causal therapy of Alzheimer’s disease (AD). In the interim, and perhaps ultimately as an adjunct to a progression inhibitor which may not be absolute, symptomatic treatment has to be optimized. Tacrine’s approval by the FDA has stimulated a search for a
