Maternal and Infant Outcomes in GCK-MODY Complicated by Pregnancy

The Journal of Clinical Endocrinology & Metabolism, 2023
Abstract Context Challenges exist in the management of Glucokinase–maturity-onset diabetes of the young (GCK-MODY), especially during pregnancy. Objective This work aimed to evaluate the prevalence of congenital anomaly in newborns ...

Variable presentations of GCK gene mutation in a family

BMJ Case Reports, 2022
We describe siblings born from non-consanguineous parents, with older sibling having asymptomatic hyperglycaemia while younger sibling presented with low birth weight and persistent hyperglycaemia from first month of life. Our case, the older sibling was heterozygous for paternally inherited GCK pathogenic variant resulting in diabetes of maturity ...

The GCK II and III subfamilies of the STE20 group kinases

Frontiers in Bioscience, 2007
The Ste20 (sterile 20) proteins are a large family of serine/threonine kinases. Since their discovery a growing body of evidence has implicated them in the regulation of signaling pathways governing cell growth, cell differentiation cell death and cell volume.

Three unreported glucokinase (GCK) missense mutations detected in the screening of thirty-two Brazilian kindreds for GCK and HNF1A-MODY

Diabetes Research and Clinical Practice, 2014
Thirty-two Brazilian families with MODY phenotype were screened for GCK and HNF1A mutations. GCK mutations were found in 8 families, all patients with mild asymptomatic hyperglycaemia; 3 of them are novel: p.Asp365Asn, p.Gly81Asp and p.Val253Leu. Previously described mutations in HNF1A were found in 2 families.

Haploinsufficiency at GCK gene is not a frequent event in MODY2 patients

Clinical Endocrinology, 2008
SummaryObjective  The aim of this study was to characterize glucokinase (GCK) alterations in maturity‐onset diabetes of the young 2 (MODY2)‐suspected patients and to investigate their clinical characteristics in relation to the parental origin of the mutation.Patients and methods  We studied a group of 57 unrelated Spanish patients presenting with ...

A new de novo mutation in the GCK gene causing MODY2

Diabetes Research and Clinical Practice, 2011
Analysis of glucokinase (GCK) gene in a 15-year-old male identified a new frameshift mutation in exon 4 caused by a heterozygous guanine deletion at position 382 (c.382delG, p.E128Xfs). No mutation was detected in the parents. Polymorphic markers' study excluded false paternity indicating that c.382delG is a novel de novo mutation.

Identifying Patients with GDM at Risk for GCK-MODY

Diabetes, 2018
GCK-MODY is characterized by stable, mild hyperglycemia without typical long-term diabetes complications. The prevalence of GCK-MODY is 0.1% in the general population. However patients are frequently misclassified as type 1, type 2, or gestational diabetes.

A RARE of hepatic Gck promoter interacts with RARα, HNF4α and COUP-TFII that affect retinoic acid- and insulin-induced Gck expression

The Journal of Nutritional Biochemistry, 2014
The expression of hepatic glucokinase gene (Gck) is regulated by hormonal and nutritional signals. How these signals integrate to regulate the hepatic Gck expression is unclear. We have shown that the hepatic Gck expression is affected by Vitamin A status and synergistically induced by insulin and retinoids in primary rat hepatocytes.
