Transcriptional Regulation of MHC Class II Gene Expression: Are MHC Class II Genes Coordinately Regulated?

MHC (Major Histocompatibility Complex) Class II antigens are polymorphic cell surface glycoproteins which bind foreign antigens and form a complex which is recognized by T lymphocyte antigen receptors. Presentation of foreign antigens by self MHC molecules to T cells triggers antigen-specific responses in the immune system.

Pemphigoid (Pg) is an autoimmune subepidermal blistering disease that affects the elderly population. The phenotype can be Bullous Pemphigoid (BP), which primarily involves the skin, or Mucous Membrane Pemphigoid (MMP), which primarily involves mucus membranes. Ocular Cicatricial Pemphigoid (OCP) and Oral Pemphigoid (OP) are subsets of MMP.

MHC class II A genes in the channel catfish (Ictalurus punctatus)

Two different cDNA sequences for major histocompatibility complex (MHC) class II beta chains from the channel catfish (Ictalurus punctatus) have been identified. Homology between these sequences and those previously identified as MHC class II B genes in other teleosts suggests they represent alleles of the DAB locus.

MHC class II deficiency patients are mutated for transcription factors that regulate the expression of major histocompatibility complex (MHC) class II genes. Four complementation groups (A-D) are defined and the gene defective in group A has been shown to encode the MHC class II transactivator (CIITA).

Combined immunodeficiency with abnormal expression of MHC class II genes

The MHC class II CID represents an example of immunodeficiency in which the defect in expression of membrane glycoproteins leads to abnormal cell to cell interactions and thus to abnormal immune responses. It represents an interesting model which confirms the importance of MHC molecules in all immune responses to foreign antigens.
