Detection of somatic copy number deletion of the CDKN2A gene by quantitative multiplex PCR for clinical practice

open access: yesFrontiers in Oncology, 2022
BackgroundA feasible method to detect somatic copy number deletion (SCND) of genes is still absent to date.MethodsInterstitial base-resolution deletion/fusion coordinates for CDKN2A were extracted from published articles and our whole genome sequencing ...

Frequent HPV-independent p16/INK4A overexpression in head and neck cancer.

open access: yesOral Oncology, 2018
OBJECTIVES p16INK4A (p16) is the most widely used clinical biomarker for Human Papillomavirus (HPV) in head and neck squamous cell cancer (HNSCC). HPV is a favourable prognostic marker in HNSCC and is used for patient stratification.

p16 gene homozygous deletions in acute lymphoblastic leukemia [PDF]

open access: yesBlood, 1995
The p16 protein is a cyclin inhibitor encoded by a gene located in 9p21, which may have antioncogenic properties, and is inactivated by homozygous p16 gene deletion or, less often, point mutation in several types of solid tumors often associated to cytogenetic evidence of 9p21 deletion.

Identification and characterization of a homozygous deletion found in ovarian ascites by representational difference analysis [PDF]

open access: yes, 1999
We have performed representational difference analysis (RDA) on DNA from tumor cells and normal fibroblasts isolated from the ascites of a patient with ovarian cancer. Five of six products of the RDA were homozygously deleted from the tumor DNA.

Ultrasound‐Activatable Piezoelectric Hydrogel Reprograms Mitochondrial Epigenetics for Osteoarthritis Therapy via the mTOR/GATD3A Axis

open access: yesAdvanced Science, EarlyView.
An ultrasound‐activatable piezoelectric hydrogel reprograms chondrocyte mitochondrial epigenetics via the mTOR/GATD3A axis, clearing damaged mitochondria and alleviating osteoarthritis progression in both mouse models and human cartilage explants. ABSTRACT The avascular nature of cartilage hinders drug delivery for osteoarthritis (OA) therapy.

Expression of p16 Within Myenteric Neurons of the Aged Colon: A Potential Marker of Declining Function

open access: yesFrontiers in Neuroscience, 2021
Human colonic neuromuscular functions decline among the elderly. The aim was to explore the involvement of senescence. A preliminary PCR study looked for age-dependent differences in expression of CDKN1A (encoding the senescence-related p21 protein) and ...

USP5 Stabilizes TGFBR1 to Drive Vascular Smooth Muscle Cell Senescence and Atherosclerosis

open access: yesAdvanced Science, EarlyView.
This study reveals that USP5 drives vascular smooth muscle cell senescence and atherosclerosis by stabilizing TGFBR1, suppressing IDH2, and promoting glycolytic reprogramming, identifying the USP5‐TGFBR1‐IDH2 axis as a potential therapeutic target. ABSTRACT Vascular smooth muscle cell (VSMC) senescence contributes importantly to atherosclerotic plaque ...

The Role of p16, p21, p27, p53 and Ki-67 Expression in the Differential Diagnosis of Cutaneous Squamous Cell Carcinomas and Keratoacanthomas: An Immunohistochemical Study

open access: yesBalkan Medical Journal, 2016
Background: Distinguishing squamous cell carcinoma (SCC) from keratoacanthoma (KA) by histopathological features may not be sufficient for a differential diagnosis, as KAs may, in some cases, imitate well-differentiated SCCs.

Bone Marrow Macrophage STING Signaling is a Direct Nexus Linking Marrow Adipose Expansion to NAD+ Depletion and Osteogenic Decline in Skeletal Aging

open access: yesAdvanced Science, EarlyView.
This study reveals that during skeletal aging, bone marrow adipocyte‐derived sDPP4 primes STING signaling in macrophages. In combination with mitochondrial DNA, this signaling activates CD38 and depletes local NAD+/NMN, thereby impairing osteogenesis.

A Dual‐Functional Nanoplatform Targeting the dsDNA–cGAS Axis to Mitigate Disc Degeneration and Aberrant Innervation [PDF]

open access: yesAdvanced science
RU@PEI‐ZIF integrates dsDNA scavenging with local cGAS inhibition to suppress the dsDNA–cGAS–STING/Netrin‐1 axis in intervertebral disc degeneration. By attenuating nucleus pulposus cell senescence, inflammatory and matrix‐catabolic responses, and pathological sensory innervation, this dual‐functional nanoplatform preserves disc integrity and reduces ...
