ESR1 methylation and ESR1 mutations in circulating tumor cells (CTCs) and paired plasma‐cfDNA of advanced breast cancer patients: A feasibility proof‐of‐concept study

open access: yesMolecular Oncology, EarlyView.
Circulating tumor cells (CTCs) and plasma cell‐free DNA (cfDNA) were analyzed to detect ESR1 mutations and methylation in patients with advanced breast cancer. CTC‐derived DNA showed higher sensitivity for mutation detection and revealed complementary genetic and epigenetic alterations, highlighting the added value of CTC analysis for understanding ...

A Computational Method for Solving $N$-Person Game

open access: yesИзвестия Иркутского государственного университета: Серия "Математика", 2017
The nonzero sum $n$-person game has been considered. It is well known that the game can be reduced to a global optimization problem [5; 7; 14]. By extending Mills’ result [5], we derive global optimality conditions for a Nash equilibrium.

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

A Global Optimality Criterion for Nonconvex Quadratic Programming over a Simplex [PDF]

open access: yes, 2005
In this paper we propose a global optimality criterion for globally minimizing a quadratic form over the standard simplex, which in addition provides a sharp lower bound for the optimal value.

Clinical performance of the urine‐based TERT promoter AbsoluteQ Digital PCR for non‐invasive detection of bladder cancer

open access: yesMolecular Oncology, EarlyView.
A urine‐based digital PCR assay targeting two hotspot TERT promoter variants detected bladder cancer with high sensitivity and no false positives in this case–control cohort. The streamlined AbsoluteQ workflow outperformed Sanger sequencing and supports non‐invasive molecular testing for bladder cancer detection.

Measuring Tax Efficiency: A Tax Optimality Index [PDF]

open access: yes
This paper introduces an index of tax optimality that measures the distance of a current tax structure from the optimal tax structure in the presence of public goods.

Intelligent Route Planning for Transport Ship Formations: A Hierarchical Global–Local Optimization and Collaborative Control Framework

open access: yesJournal of Marine Science and Engineering
Multi-vessel formation shipping demonstrates significant potential for enhancing maritime transportation efficiency and economy. However, existing route planning systems inadequately address the unique challenges of formations, where traditional methods ...

Developmental programmes drive cellular plasticity, disease progression and therapy resistance in lung adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.

Non-convex quadratic minimization problems with quadratic constraints: Global optimality conditions

open access: yes, 2007
In this paper, we first examine how global optimality of non-convex constrained optimization problems is related to Lagrange multiplier conditions. We then establish Lagrange multiplier conditions for global optimality of general quadratic minimization ...

CAGE: A Curiosity-Driven Graph-Based Explore-Exploit Algorithm for Solving Deterministic Environment MDPs With Limited Episode Problem

open access: yesIEEE Access
The explore-exploit dilemma in Markov Decision Processes (MDPs) is a fundamental challenge, especially in deterministic environments akin to real-world scenarios.
