A genetically encoded, imaging‐based reporter enables direct measurement of O‐GlcNAc transferase (OGT) activity in living cells. The system exploits OGT‐dependent proteolysis of HCF‐1 to drive nuclear and cytoplasmic redistribution of fluorescent signals, allowing cell‐by‐cell spatially resolved quantification of OGT activity.
