Systematic unravelling of the inulin hydrolase from Bacillus amyloliquefaciens for efficient conversion of inulin to poly-(γ-glutamic acid)

open access: yesBiotechnology for Biofuels, 2019
Background Bacillus amyloliquefaciens NB is a newly discovered strain, which produces poly-(γ-glutamic acid) (γ-PGA) from raw extracted inulin of Jerusalem artichoke tubers; however, the underlying mechanisms remain unknown.

Structural and functional insights into extreme thermal stability and activity of two GH12 domains of a multidomain glycosidase from a hyperthermophilic euryarchaeon

open access: yesThe FEBS Journal, EarlyView.
The multidomain glycosidase MDG from the hyperthermophilic euryarchaeon Thermococcus sp. contains five protein domains: three catalytic domains (GH5, GH12‐1, and GH12‐2) and two cellulose‐binding modules (CBM2). In this study, the GH12‐1 and GH12‐2 domains were individually purified and biochemically characterized.

Immobilization of Glycoside Hydrolase Families GH1, GH13, and GH70: State of the Art and Perspectives

open access: yesMolecules, 2016
Glycoside hydrolases (GH) are enzymes capable to hydrolyze the glycosidic bond between two carbohydrates or even between a carbohydrate and a non-carbohydrate moiety.

Electrostatic potential as a reactivity scoring function in computer‐assisted enzyme engineering

open access: yesThe FEBS Journal, EarlyView.
We present a computational metric that quantifies the electrostatic complementarity of enzymes to the charge distribution of substrates at the transition state. Implemented in BindScan, this metric accurately predicts functionally relevant mutations in glycoside hydrolases.

Catalytic Mechanism of a Novel Glycoside Hydrolase Family 16 “Elongating” β-Transglycosylase*

open access: yesJournal of Biological Chemistry, 2016
Carbohydrates are complex macromolecules in biological metabolism. Enzymatic synthesis of carbohydrates is recognized as a powerful tool to overcome the problems associated with large scale synthesis of carbohydrates.

Anions and citrate inhibit LsAA9A, a lytic polysaccharide monooxygenase (LPMO)

open access: yesThe FEBS Journal, EarlyView.
The fungal lytic polysaccharide monooxygenase LsAA9A binds chloride in its crystallization mother liquor, leading to LsAA9A inhibition. Other halides except for fluoride inhibit LsAA9A activity at high concentrations. Transferring crystals to a different solution with lower ionic strength leads to binding of citrate buffer, leading also to the ...

Molecular characterization of a family 5 glycoside hydrolase suggests an induced-fit enzymatic mechanism

open access: yesScientific Reports, 2016
Glycoside hydrolases (GHs) play fundamental roles in the decomposition of lignocellulosic biomaterials. Here, we report the full-length structure of a cellulase from Bacillus licheniformis (BlCel5B), a member of the GH5 subfamily 4 that is entirely ...
