Molecular biology of glutathione peroxidase 4: from genomic structure to developmental expression and neural function [PDF]

open access: yes, 2017
Selenoproteins have been recognized as modulators of brain function and signaling. Phospholipid hydroperoxide glutathione peroxidase (GPx4/PHGPx) is a unique member of the selenium-dependent glutathione peroxidases in mammals with a pivotal role in brain

Reduced selenium concentrations and glutathione peroxidase activity in preeclamptic pregnancies [PDF]

open access: yes, 2008
Preeclampsia is a pregnancy-specific condition affecting 2-7% of women and a leading cause of perinatal and maternal morbidity and mortality. Preeclampsia may also predispose the fetus to increased risks of adult cardiovascular disease.

HPD is an m6A Methyltransferase that Protects Colorectal Cancer Cells from Ferroptotic Cell Death by m6A Methylating SLC7A11/GPX4

open access: yesAdvanced Science, EarlyView.
This study reveals that the tyrosine metabolic enzyme HPD functions as a previously uncharacterized, METTL3‐independent m6A methyltransferase. It promotes colorectal tumor progression by coordinately regulating the SLC7A11/GPX4 axis to suppress ferroptosis.

Gallium‐Doped MXene Nanozymes Protect Liver Through Multi‐Death Pathway Blockade and Hepatocyte Regeneration

open access: yesAdvanced Science, EarlyView.
This study develops gallium‐doped V2C MXene nanozymes (Ga‐V2C) to treat acetaminophen‐induced liver injury through multi‐death pathway blockade and hepatocyte regeneration. Unlike conventional single‐target therapies like N‐acetylcysteine, Ga‐V2C nanozymes enable oxidative stress suppression, apoptosis, and ferroptosis inhibition, and enhanced ...

HMGB1 mediates lipopolysaccharide-induced inflammation via interacting with GPX4 in colon cancer cells

open access: yesCancer Cell International, 2020
Background Inflammation is one of a main reason for colon cancer progression and poor prognosis. The high-mobility group box-1 (HMGB1) and glutathione peroxidase 4 (GPX4) are responsible for inflammation, but the relationship between HMGB1 and GPX4 ...

TRIM56 Aggravates Cerebral Ischemia‐Reperfusion Injury via Inhibiting KLF4‐Activated Ferroptosis Signaling

open access: yesAdvanced Science, EarlyView.
This study reveals that the E3 ubiquitin ligase TRIM56 exacerbates neuronal ferroptosis and brain damage by mediating K48‐linked ubiquitination and degradation of KLF4, leading to suppression of the xCT/GSH/GPX4 axis. Targeting TRIM56 alleviates cerebral ischemia‐reperfusion injury in vivo and in vitro, highlighting its therapeutic potential.

Genetic polymorphism in selenoprotein P modifies the response to selenium-rich foods on blood levels of selenium and selenoprotein P in a randomized dietary intervention study in Danes [PDF]

open access: yes, 2018
Background Selenium is an essential trace element and is suggested to play a role in the etiology of a number of chronic diseases. Genetic variation in genes encoding selenoproteins, such as selenoprotein P and the glutathione peroxidases, may affect ...

LncRNA-PVT1 Inhibits Ferroptosis through Activating STAT3/GPX4 Axis to Promote Osteosarcoma Progression

open access: yesFrontiers in Bioscience-Landmark
Background: Osteosarcoma (OS) is a primary malignant bone tumor in the pediatric and adolescent populations. Long non-coding RNAs (LncRNAs), such as plasma-cytoma variant translocation 1 (PVT1), have emerged as significant regulators of OS metastasis ...

Effects of gpx4 Haploid Insufficiency on GPx4 Activity, Selenium Concentration, and Paraquat-Induced Protein Oxidation in Murine Tissues

open access: yesExperimental Biology and Medicine, 2005
Selenium-dependent glutathione peroxidase-4 (GPx4) catalyzes the reduction of phospholipid hydroperoxides. Because a full gpx4 knockout is embryonic lethal, we examined the effect of deletion of one copy of gpx4 on the activities of three selenoperoxidases (GPx1, GPx3, and GPx4), selenium concentrations, and pro-oxidant-induced protein oxidation in ...

Investigating Mitochondrial Viscosity in Ferroptosis‐Mediated Drug‐Induced Liver Injury using a Double‐Targeted Strategy

open access: yesAdvanced Science, EarlyView.
Utilizing a mitochondrial double‐targeted strategy, a novel near‐infrared fluorescent probe (FTZ‐2) is established for monitoring viscosity variations in ferroptosis‐mediated drug‐induced liver injury. In summary, this work provided a practical strategy for the early diagnosis and evaluation of liver injury.
