Induction of IL‐9‐producing CD8+ T cells by ascochlorin derivatives

A newly synthesised ascochlorin derivative, N184, induces small amounts of IL‐9‐producing CD8+ T cells. N184 improved the survival of CD8+ T cells and enhanced their antitumour activity, partially in an IL‐9‐dependent manner. The antitumour effect was completely dependent on IFNγ secretion rather than the cytotoxic activity of CD8+ T cells.

Opioids in breast cancer: Between analgesia and modulation of tumour progression

Preclinical investigations consistently demonstrate that activation of μ‐opioid receptors and δ‐opioid receptors promote proliferation, migration, angiogenesis, epithelial‐mesenchymal transition, acquisition of cancer stem cell phenotypes, and chemoresistance.

Abstract In the 2025 novel drug mini‐review, one can take a full measure of the ingenuity that underlies current drug design and development, despite the year's smaller harvest (46 novel drugs) compared to 2024 (53) and 2023 (70). 54% of the novel drugs are first‐in‐class (FIC).

TGF‐β Inhibitor Potentiates Osimertinib‐Induced Anti‐Tumor Immunity in Egfr‐Mutant Lung Cancer

Osimertinib enhances CD8+ T cell activity but also upregulates immunosuppressive TGF‐β in Egfr‐mutant lung tumors. This study demonstrates that TGF‐β inhibition with nintedanib or vactosertib potentiates osimertinib‐induced CD8+ T cell–mediated immunity and reduce CD206+ macrophages. CD8+ T Cell Infiltration Elicits Molecular Subtype‐Biased Clinical Outcomes in Gastric Cancer Patients

CD8+ T cell infiltration demonstrates distinct clinical significance across molecular subtypes of gastric cancer. High infiltration correlates with improved survival in EBV‐positive and CIN subtypes, while low infiltration predicts better adjuvant chemotherapy response in MSI patients.
