Identification of novel small molecule inhibitors of ETS transcription factors
ETS transcription factors play an essential role in tumourigenesis and are indispensable for sprouting angiogenesis, a hallmark of cancer, which fuels tumour expansion and dissemination. Thus, targeting ETS transcription factor function could represent an effective, multifaceted strategy to block tumour growth. The evolutionarily conserved E‐Twenty‐Six

A Comparative Study of Disordered and Ordered Protein Folding Dynamics Using Computational Simulation [PDF]
Folding protein dynamics has been an area of high interest for quite some time, especially given the increased focus on the field of Biophysics. Because folding dynamics occur on such short time scales, empirical techniques developed for more "static" protein events, such as X-ray crystallography, nuclear magnetic resonance, and green fluorescent ...

Development of a Multiphoton Fluorescence Lifetime Imaging Microscopy (FLIM) system using a Streak Camera [PDF]
We report the development and detailed calibration of a multiphoton fluorescence lifetime imaging system (FLIM) using a streak camera. The present system is versatile with high spatial (0.2 micron) and temporal (50 psec) resolution and allows rapid data acquisition and reliable and reproducible lifetime determinations.

The Saccharomyces cerevisiae amino acid transporter Lyp1 has a broad substrate spectrum
In Saccharomyces cerevisiae, Yeast Amino acid Transporter family members mediate the import of amino acids, ranging from substrate specialists to generalists. Here, we show that the specialist transporter, Lyp1, has a broader substrate spectrum than previously described, with affinity constants spanning from micromolar to millimolar.

Evaluation of chemical fluorescent dyes as a protein conjugation partner for live cell imaging.
To optimize live cell fluorescence imaging, the choice of fluorescent substrate is a critical factor. Although genetically encoded fluorescent proteins have been used widely, chemical fluorescent dyes are still useful when conjugated to proteins or ...

Cancer‐associated fibroblasts (CAFs) promote cancer growth, invasion (metastasis), and drug resistance. Here, we identified functional and diverse circulating CAFs (cCAFs) in patients with metastatic prostate cancer (mPCa). cCAFs were found in higher numbers and were functional and diverse in mPCa patients versus healthy individuals, suggesting their ...

A comparative study of circulating tumor cell isolation and enumeration technologies in lung cancer
Lung cancer cells were spiked into donor blood to evaluate the recovery rates of the following circulating tumor cell (CTC) enrichment technologies: CellMag™, EasySep™, RosetteSep™, Parsortix® PR1, and Parsortix® Prototype systems. Each method's advantages and disadvantages are described.

Photoactivatable Green Fluorescent Protein-Tubulin [PDF]
Direct observations of live cells expressing fluorescently tagged tubulin have led to important advances in our understanding of mitosis. A limitation of this approach is that all of the cells' microtubules are fluorescent and thus observation of the behavior of specific subsets of microtubules is precluded.

A Dark Excited State of Fluorescent Protein Chromophores, Considered as Brooker Dyes [PDF]
The green fluorescent protein (GFP) chromophore is an asymmetric monomethine dye system. In the resonance color theory of dyes, a strong optical excitation arises from interactions of two valence-bond structures with a third, higher structure. We use correlated quantum chemistry to show that the anionic chromophore is a resonant Brooker dye, and that ...
