Dual targeting of RET and SRC synergizes in RET fusion‐positive cancer cells

open access: yesMolecular Oncology, EarlyView.
Despite the strong activity of selective RET tyrosine kinase inhibitors (TKIs), resistance of RET fusion‐positive (RET+) lung cancer and thyroid cancer frequently occurs and is mainly driven by RET‐independent bypass mechanisms. Son et al. show that SRC TKIs significantly inhibit PAK and AKT survival signaling and enhance the efficacy of RET TKIs in ...

53BP1 can limit sister-chromatid rupture and rearrangements driven by a distinct ultrafine DNA bridging-breakage process

open access: yesNature Communications, 2018
Chromosome instability is associated with cancer formation. Here the authors identify in cultured human cancer cells a non-canonical DNA bridge breakage pathway leading to chromosome missegregation and rearrangements triggered by sister DNA ...

Chaotic micromixing in open wells using audio-frequency acoustic microstreaming

open access: yesBioTechniques, 2009
Mixing fluids for biochemical assays is problematic when volumes are very small (on the order of the 10 µL typical of single drops), which has inspired the development of many micromixing devices.

Detection of circulating tumor DNA in colorectal cancer patients using a methylation‐specific droplet digital PCR multiplex

open access: yesMolecular Oncology, EarlyView.
We developed a cost‐effective methylation‐specific droplet digital PCR multiplex assay containing tissue‐conserved and tumor‐specific methylation markers. The assay can detect circulating tumor DNA with high accuracy in patients with localized and metastatic colorectal cancer.

Development and implementation of a gyrolab-based generic anti-drug antibody assay for antibody-drug conjugates in cynomolgus monkey studies

open access: yesFrontiers in Immunology
IntroductionNon-clinical immunogenicity yields valuable insights into pharmacokinetics, efficacy, and safety. Given the complex nature of ADC therapeutics, early detection of anti-drug antibodies (ADAs) is critical for elucidating exposure and toxicity ...

Perspectives in educating molecular pathologists on liquid biopsy: Toward integrative, equitable, and decentralized precision oncology

open access: yesMolecular Oncology, EarlyView.
Liquid biopsy enables minimally invasive, real‐time molecular profiling through analysis of circulating biomarkers in biological fluids. This Perspective highlights the importance of training pathologists through integrative educational programs, such as the European Masters in Molecular Pathology, to ensure effective and equitable implementation of ...

Flutter to tumble transition of buoyant spheres triggered by rotational inertia changes

open access: yesNature Communications, 2018
Spiral and zigzag trajectories of rising bubbles and falling leaves represent some of the path instabilities commonly observed for objects moving through fluids. Here, Mathai et al.

Mechanisms of transcriptional regulation of ecdysone response

open access: yesВавиловский журнал генетики и селекции, 2019
The mechanisms of ecdysone-dependent expression have been studied for many decades. Initially, the activation of individual genes under the influence of ecdysone was studied on the model of polythene chromosomes from salivary glands of Drosophila ...

Transcriptional network analysis of PTEN‐protein‐deficient prostate tumors reveals robust stromal reprogramming and signs of senescent paracrine communication

open access: yesMolecular Oncology, EarlyView.
Combining PTEN protein assessment and transcriptomic profiling of prostate tumors, we uncovered a network enriched in senescence and extracellular matrix (ECM) programs associated with PTEN loss and conserved in a mouse model. We show that PTEN‐deficient cells trigger paracrine remodeling of the surrounding stroma and this information could help ...
