Targeting GSDME-mediated macrophage polarization for enhanced antitumor immunity in hepatocellular carcinoma

Cellular and Molecular Immunology
Despite the notable efficacy of anti-PD1 therapy in the management of hepatocellular carcinoma (HCC) patients, resistance in most individuals necessitates additional investigation. For this study, we collected tumor tissues from nine HCC patients receiving anti-PD1 monotherapy and conducted RNA sequencing.

GSDME-mediated pyroptosis contributes to chemotherapy-induced platelet hyperactivity and thrombotic potential

Blood
Abstract Thrombotic complications due to platelet hyperreactivity are a major cause of death in patients undergoing chemotherapy. However, the underlying mechanisms are not fully understood. Herein, using human platelets and platelets from mice lacking gasdermin E (GSDME), we show that GSDME is functionally ...

GSDME unlocks astrocyte-driven neurotoxicity in Alzheimer’s Disease

SUMMARY Astrocytic calcium dysregulation and reactivity precede Aβ deposition in amyloid-β deposition in Alzheimer’s disease (AD) but the neurotoxic mechanisms remain unclear. We show that GSDME acts as a switch, linking MAM-mediated calcium release to astrocyte-driven neurotoxicity.

Alpinumisoflavone triggers GSDME-dependent pyroptosis in esophageal squamous cell carcinomas.

Anatomical record (Hoboken, N.J. : 2007), 2021
Esophageal squamous cell carcinoma (ESCC) presents a common human malignancy in the digestive system. We aimed to explore the critical effects of alpinumisoflavone (AIF) on ESCC in vitro and in vivo. The cell counting kit-8 assay was used to determine cell viability.

Euphohelioscopin A enhances NK cell antitumor immunity through GSDME-triggered pyroptosis

Journal of Leukocyte Biology
Abstract Immune evasion by cancer cells poses a significant challenge for natural killer cell–based immunotherapy. Pyroptosis, a newly discovered form of programmed cell death, has shown great potential for enhancing the antitumor immunity of natural killer cells.

CASP3-GSDME-mediated trophoblast pyroptosis contributes to systemic inflammation

Abstract Early-onset preeclampsia (EOPE) is associated with excessive apoptosis and inflammation, but the mechanistic link between these processes remains enigma. Here, we report elevated circulating pro-apoptotic proteins in EOPE patients at early pregnancy, along with concurrent CASP3 activation and GSDME cleavage in a subset of EOPE ...

High glucose promotes lipopolysaccharide‐induced macrophage pyroptosis through GSDME O‐GlcNAcylation

Journal of Periodontal Research
AbstractAimThe high glucose (HG) environment in diabetic periodontitis aggravates the damage of periodontal tissue. Pyroptosis has been shown to be positively correlated with the severity of periodontitis, including macrophage pyroptosis. O‐GlcNAcylation is a posttranslational modification that is involved in the pathogenesis of periodontitis. However,

Inhibiting apoptosis and GSDME-mediated pyroptosis attenuates hepatic injury in septic mice

Archives of Biochemistry and Biophysics
Sepsis is characterized by severe inflammation and organ dysfunction resulting from a dysregulated organismal response to infection. Although pyroptosis has been presumably shown to be a major cause of multiple organ failure and septic death, whether gasdermin E (GSDME)-mediated pyroptosis occurs in septic liver injury and whether inhibiting apoptosis ...
