Assessment of HDACi-Induced Protein Cleavage by Caspases

2016
Aberrant histone deacetylase (HDAC) activity often correlates with neoplastic transformation and inhibition of HDACs by small molecules has emerged as a promising strategy to treat hematological malignancies in particular. Treatment with HDAC inhibitors (HDACis) often prompts tumor cells to undergo apoptosis, thereby causing a caspase-dependent ...

Colony Formation Assay to Test the Impact of HDACi on Leukemic Cells

2022
One of the main characteristics of cancer is the rapid proliferation of transformed cells. Cancer therapies aim to kill such cells. Cancer clones surviving therapy can be resistant to the treatment, but they can also lose the ability to proliferate. The ability of single cells to proliferate can be monitored in vitro and can provide insights into the ...

Encapsulation of the HDACi Ex527 into Liposomes and Polymer-Based Particles

2016
Incorporation of drugs into particles can improve their therapeutic effectiveness. Solubility, half-life time, targeting, and the release of the drug can be modified by the encapsulation into a particle. Histone deacetylase inhibitors have a great potential to be used as therapeutics for many different diseases.

PEGylated Poly‐HDACi: A Designer Polyprodrug from Optimized Drug Units**

Chemistry – A European Journal, 2021
AbstractWe designed, synthesized, and characterized a tri‐block copolymer. Its hydrophobic part, a chain of histone deacetylase inhibitor (HDACi) prodrug, was symmetrically flanked by two identical PEG blocks, whereas the built‐in HDACi was a linear molecule, terminated with a thiol at one end, and a hydroxyl group at the other.

HDACi: molecular mechanisms and therapeutic implications in the innate immune system

Immunology & Cell Biology, 2011
Histone deacetylase inhibitors (HDACi) are an emerging class of novel anti‐cancer drugs that cause growth arrest, differentiation and apoptosis of tumor cells. In addition, many advances have been made in understanding the immunoregulation of Toll‐like receptors, NOD‐like receptors and interferons that have recently generated new momentum for the study

Analysis of HDACi-Induced Changes in Chromosomal Passenger Complex Localization

2016
The chromosomal passenger complex (CPC) is a key regulator of cell division. Its proper localization during the different phases of mitosis and cytokinesis is crucial for the exertion of its various functions. HDACi treatment has been demonstrated to disturb the centromeric localization of the CPC in tumor cells, thus leading to severe mitotic defects ...

Abstract 1146: Multiple HDAC inhibitors (HDACi) reduce the expression of the oncogenic miR-17∼92 cluster; a pan HDACi class effect

Cancer Research, 2011
Abstract miR-17∼92 is an oncogenic polycistronic cluster encoding six mature miRNAs (MIR-17, MIR-20A, MIR-18A, MIR-19A, MIR-19B, and MIR-92A). This oncogenic cluster is driven by MYC activity and is highly-expressed in many tumor types where it has been shown to promote tumorigenesis through the suppression of oncogene-induced senescence.

Assessment of HDACi-Induced Acetylation of Nonhistone Proteins by Mass Spectrometry

2016
Posttranslational acetylation of lysine residues has been discovered as multifaceted regulatory modification for various nuclear, cytoplasmic, and mitochondrial proteins. The implementation of high-resolution and high-throughput mass spectrometry (MS) approaches has led to the identification of a hitherto underappreciated, large number of acetylation ...

Abstract 2557: Preclinical synergy of Plk1 inhibitors and HDACIs

Cancer Research, 2011
Abstract Our laboratory is interested in the synergy between spindle check point inhibitors and histone deacetylase inhibitors (HDACIs). Previous work investigated two polo-like kinase inhibitors in prostate cancer and in non-small cell lung cancer (NSCLC).
