Thermal Shifts Dominate Single‐Gene Responses to Transcription‐Targeting Antibiotics

open access: yesAdvanced Science, EarlyView.
Thermal shifts reshape bacterial responses to transcription‐targeting antibiotics. In E. coli, temperature can dominate antibiotic‐induced genome‐wide expression, particularly outside 30°C–39°C, through changes in adenosine triphosphate (ATP), nucleoid organization, and drug‐target engagement.

AI‐Designed TREM1‐Targeted LYTAC Nanoparticles Reprogram the Neuroimmune Microenvironment in Traumatic Brain Injury

open access: yesAdvanced Science, EarlyView.
AI‐enabled de novo design of a TREM1 binder, integrated into a BBB‐crossing and macrophage‐targeted nanoplatform, enables TREM1 degradation and attenuates neuroinflammation, thereby promoting neuroprotection and tissue repair after traumatic brain injury. ABSTRACT Secondary neuroinflammation drives progressive damage after traumatic brain injury (TBI),

Neonatal Sevoflurane Exposure Induces Long‐Term Cognitive Impairment via Epigenetically Mediated MMP9 Activation and Perineuronal Net Disruption in the Hippocampal CA2

open access: yesAdvanced Science, EarlyView.
Hippocampal CA2 perineuronal nets ensheathing excitatory pyramidal neurons are selectively vulnerable to repeated neonatal sevoflurane exposure. Epigenetic upregulation of MMP9 via reduced H3K27me3 disrupts PNN integrity, impairs BDNF–TrkB signaling, and causes synaptic protein loss and dendritic spine deficits, leading to persistent social and spatial

Early‐Life Manganese Overexposure Activates Microglial Phagocytosis through the YY1/TREM2 Axis Leading to Hippocampal Synaptic Remodeling in Mice

open access: yesAdvanced Science, EarlyView.
Early‐life manganese exposure induces behavioral deficits and aberrant hippocampal synaptic remodeling by enhancing microglial phagocytosis in young adult male mice. Mechanistically, manganese promotes Yin Yang 1 (YY1)/triggering receptor expressed on myeloid cells 2 (TREM2) signaling through enhanced HIF1α‐mediated transcription and suppressed SMURF2 ...

Decoding Biomolecular Phase Separation: From Physiological Roles to Pathological Mechanisms and Therapeutic Biomaterials Design

open access: yesAdvanced Science, EarlyView.
This review systematically elaborates on the mechanisms underlying physiological regulation and pathological dysregulation of biomolecular phase separation. It also summarizes advances in phase‑separation‑targeted therapeutic strategies and the design of functional biomaterials inspired by this process.

EphB1 Receptor Blockade Augments Morphine Antinociception via Inhibiting GRK2‐Mediated µ‐Opioid Receptor Internalization

open access: yesAdvanced Science, EarlyView.
Administration of opioids activates ephrinB2‐EphB1 receptor signaling. Activated EphB1 receptors induce GRK2‐mediated phosphorylation of MOR at Ser375, which initiates the internalization of MOR and reduces opioid antinociception at the behavioral level. ABSTRACT Opioids are the primary treatment for severe pain in clinical practice.

Sonogenetic Control of Engineered Probiotics for the Treatment of MASH by Modulating the Gut‐Liver Axis

open access: yesAdvanced Science, EarlyView.
Researchers developed an engineered probiotic encapsulated in an inulin hydrogel, enabling ultrasound‐triggered intestinal delivery of IL‐22. In MASH model mice, this system remodeled gut microbiota, strengthened the intestinal barrier and reduced portal LPS and hepatic IL‐6.

Cellular Mechanisms Enabling Mitochondria Transfer and Transplantation

open access: yesAdvanced Science, EarlyView.
Mitochondrial transfer and transplantation (mitoTT) is examined as a multistep process governed by distinct transfer routes and post‐transfer checkpoints. Mechanisms controlling mitochondrial uptake, network integration, cristae remodeling, mtDNA maintenance, immune sensing, and quality control are highlighted, revealing key biological and ...
