Cancer‐associated fibroblasts (CAFs) are the major component of the tumor stroma. This review will highlight the CAF subtypes within the tumor microenvironment and their role in tumor progression. PRRX2 is highly expressed in pancreatic cancer and correlates with patient prognosis. PRRX2 promotes pancreatic cancer cell proliferation, invasion, and metastasis, and inhibits pancreatic cancer cell senescence. PRRX2 transcriptionally regulates GLI2 and activates the Hedgehog pathway.

Trio exome sequencing allowed the identification of two novel compound heterozygous variants in PIK3C2A, defining the fifth family presenting a PIK3C2A‐related syndrome characterized by pulverulent cataracts and deafness. Functional testing revealed impaired PI metabolism and primary dysfunction phenotype.
