Giant basal cell carcinoma of anterior chest wall reveals metastasis to lungs: A case report
Basal cell carcinoma (BCC) is the most common cutaneous malignancy in the world, and the incidence of pulmonary metastasis is exceedingly rare. We present a case of middle‐aged male with findings consistent with BCC with metastasis to the lungs managed ...

Targeted therapy was evaluated in SHH medulloblastoma using neuroepithelial stem cell (NES) and tumor‐derived NES‐like (tNES) models in 2D monolayers and 3D spheroids. PI3K, AKT, and CDK4/6 inhibitors had minimal effects in NES but markedly reduced viability and growth and induced apoptosis in tNES cells, revealing distinct therapeutic vulnerabilities.

Natural Biomaterials for Osteochondral Repair: From Source to Strategy
Biological origin‐guided overview of natural biomaterials and therapeutic strategies for osteochondral tissue engineering. The circular diagram categorizes representative materials and strategies into plant/algae‐derived, microbial‐derived, animal‐derived, and human‐derived sources, centered on an osteochondral defect repair model.

Hedgehog pathway as a drug target: Smoothened inhibitors in development
Emerging laboratory and clinical investigations demonstrate that Hedgehog signaling (Hh) represents a novel therapeutic target in various human cancers. This conserved signaling pathway precisely regulates self-renewal and terminal differentiation in embryonic development, but is typically silenced in adult tissues, with reactivation usually only ...

In fibrotic distal lung regions, CD66c+ basal cells emerge as a pathological state. Using human distal lung organoids, this study identifies CD66c+ basal cells as a pro‐fibrotic state arising through transdifferentiation from secretory, AT2, and basal cells.

Development of posaconazole-based analogues as hedgehog signaling pathway inhibitors [PDF]
Inhibition of the hedgehog (Hh) signaling pathway has been validated as a therapeutic strategy to treat basal cell carcinoma and holds potential for several other forms of human cancer. Itraconazole and posaconazole are clinically useful triazole anti-fungals that are being repurposed as anti-cancer agents based on their ability to inhibit the Hh ...

Pancreatic neuroendocrine tumors frequently silence MEN1 through epigenetic mechanisms. Here, SIRT7 recruits DNMT1 to the MEN1 promoter, drives hypermethylation, and enhances DNA repair. Inhibiting SIRT7 restores MEN1, reduces MRN complex abundance, impairs double‐strand break repair, and sensitizes PanNET models to radiation, supporting SIRT7 as a ...

Single‐cell RNA sequencing verified the presence of the VIM+ biliary epithelial cell (BEC) in the bile ducts of NAS patients. The hypoxia/TGF‐β‐CREM‐VIM axis mediated phenotypic switch toward VIM+ BEC is validated using a hypoxia/TGF‐β‐stimulated cellular model, a rat liver transplantation model, BEC‐specific Crem conditional knockout rats, and BEC ...
