An Accessible Microfluidic Platform for the Generation of Polarized Brain Organoids

open access: yesAdvanced Materials Technologies, EarlyView.
This study presents an accessible double‐gradient microfluidic device that enables controlled morphogen delivery and spatially polarized differentiation in brain organoids. By integrating a pump‐free design and stable fabrication method, the platform generates dorsal–ventral neuronal asymmetry, offering a reproducible and versatile approach to model ...

Topological Analysis of Hedgehog Acyltransferase, a Multipalmitoylated Transmembrane Protein [PDF]

open access: yesJournal of Biological Chemistry, 2015
Hedgehog proteins are secreted morphogens that play critical roles in development and disease. During maturation of the proteins through the secretory pathway, they are modified by the addition of N-terminal palmitic acid and C-terminal cholesterol moieties, both of which are critical for their correct function and localization.

Engineering Osteoimmune Responses with Functionalized Orthopedic Implants for Post‐Operative Osteosarcoma Treatment

open access: yesAdvanced Science, EarlyView.
Osteosarcoma is the most common primary bone tumor with limited treatment options and a terrible prognosis. This review provides a comprehensive summary of the recent development of osteoimmunomodulatory implants for post‐operative osteosarcoma treatment, of which the potential utility in evoking durable anti‐osteosarcoma immunity and accelerating bone

Restriction of Individual Branched‐Chain Amino Acids has Distinct Effects on the Development and Progression of Alzheimer's Disease in 3xTg Mice

open access: yesAdvanced Science, EarlyView.
Protein restriction (PR) slows Alzheimer's disease (AD) in mice, and other benefits of PR are due to decreased branched‐chain amino acids (BCAAs). We show that restricting any BCAA has benefits, with sex‐ and BCAA‐specific impacts on pathology, molecular signaling, and cognition.

Size‐Modulated Mesoderm‐Endoderm Divergence and Myocardial Cavitation in Micropatterned Cardioids

open access: yesAdvanced Science, EarlyView.
Micropatterned cardioids, CRISPR‐engineered reporter hiPSCs, deep‐tissue imaging, and single‐cell RNA sequencing are integrated to model mesoderm‐endoderm co‐development. Heart‐foregut crosstalk promotes single large cavitation inside cardioids, resembling early heart chamber formation. ABSTRACT The human heart, originating from the splanchnic mesoderm,

Sonic hedgehog is basolaterally sorted from the TGN and transcytosed to the apical domain involving Dispatched-1 at Rab11-ARE

open access: yesFrontiers in Cell and Developmental Biology, 2022
Hedgehog proteins (Hhs) secretion from apical and/or basolateral domains occurs in different epithelial cells impacting development and tissue homeostasis.

Phosphoinositides Regulate Ciliary Protein Trafficking to Modulate Hedgehog Signaling [PDF]

open access: yesDevelopmental Cell, 2015
Primary cilia interpret vertebrate Hedgehog (Hh) signals. Why cilia are essential for signaling is unclear. One possibility is that some forms of signaling require a distinct membrane lipid composition, found at cilia. We found that the ciliary membrane contains a particular phosphoinositide, PI(4)P, whereas a different phosphoinositide, PI(4,5)P2, is ...
