CNOT1 cooperates with LMNA to aggravate osteosarcoma tumorigenesis through the Hedgehog signaling pathway

open access: yesMolecular Oncology, 2017
While treatments for childhood osteosarcoma have improved, the overall survival for this common type of bone cancer has not changed for three decades, and thus, new targets for therapeutic development are needed.

Emodin Alleviates Sepsis‐Induced Multiorgan Damage by Inhibiting NETosis through Targeting Neutrophils BCL‐10

open access: yesAdvanced Science, EarlyView.
Emodin targets BCL‐10 to modulate the BCL‐10/MALT1 complex, thereby suppressing NF‐κB activation and significantly exerting multiorgan protective effects in sepsis. Abstract Sepsis is a life‐threatening condition caused by dysregulated host responses to infection, characterized by excessive inflammation and abnormal coagulation.

Nrf2 Drives Epigenetic Reprogramming and Acts as the Master Regulator of KLF4 Expression and Activity in Arsenic‐Induced Transformation

open access: yesAdvanced Science, EarlyView.
Nrf2 isn't just a protector—it's a driver of cancer stemness. The study uncovers how arsenic‐activated Nrf2 directly upregulates KLF4, a key pluripotency factor, fueling oncogenic reprogramming. Through enhancer activation and self‐reinforcing loop, Nrf2 and KLF4 co‐opt gene networks linked to epithelial‐to‐mesenchymal transition and tumor growth ...

Hedgehog Signal and Genetic Disorders

open access: yesFrontiers in Genetics, 2019
The hedgehog (Hh) family comprises sonic hedgehog (Shh), Indian hedgehog (Ihh), and desert hedgehog (Dhh), which are versatile signaling molecules involved in a wide spectrum of biological events including cell differentiation, proliferation, and ...

Single‐Cell Analysis Reveals that Vitamin C Inhibits Bone Metastasis of Renal Cancer via Cell Cycle Arrest and Microenvironment Remodeling

open access: yesAdvanced Science, EarlyView.
3D‐cultured renal cancer cells exhibit stem‐like phenotypes and are more efficient in generating bone metastases. Vitamin C treatment can suppress the development of bone metastasis. Single‐cell transcriptomic analysis reveals that vitamin C inhibits metastasis by inducing tumor cell cycle arrest, suppressing osteoclast differentiation, and reducing ...

Genes targeted by the Hedgehog-signaling pathway can be regulated by Estrogen related receptor β

open access: yesBMC Molecular Biology, 2015
BackgroundNuclear receptor family member, Estrogen related receptor β, and the Hedgehog signal transduction pathway are both reported to relate to tumorigenesis and induced pluripotent stem cell reprogramming.

RPL6 Interacts with HMGCS1 to Stabilize HIF‐1α by Promoting Cholesterol Production in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
The study found that RPL6 is frequently upregulated in HCC tissues with extrahepatic metastasis. Elevated RPL6 bound to and increased the stability of HMGCS1 mRNA, thereby enhancing intracellular cholesterol levels. Subsequently, increased cholesterol promotes cancer metastasis in HCC by stabilizing HIF‐1α protein.

The Proteasome and Cul3-Dependent Protein Ubiquitination Is Required for Gli Protein-Mediated Activation of Gene Expression in the Hedgehog Pathway

open access: yesCells
Many cellular processes are regulated by proteasome-mediated protein degradation, including regulation of signaling pathways and gene expression. Among the pathways regulated by the ubiquitin–proteasome system is the Hedgehog pathway and its downstream ...
