In fibrotic distal lung regions, CD66c+ basal cells emerge as a pathological state. Using human distal lung organoids, this study identifies CD66c+ basal cells as a pro‐fibrotic state arising through transdifferentiation from secretory, AT2, and basal cells.

Ethanol Effects on Early Developmental Stages Studied Using the Zebrafish
Fetal alcohol spectrum disorder (FASD) results from prenatal ethanol exposure. The zebrafish (Danio rerio) is an outstanding in vivo FASD model. Early development produced the three germ layers and embryonic axes patterning.

Pancreatic neuroendocrine tumors frequently silence MEN1 through epigenetic mechanisms. Here, SIRT7 recruits DNMT1 to the MEN1 promoter, drives hypermethylation, and enhances DNA repair. Inhibiting SIRT7 restores MEN1, reduces MRN complex abundance, impairs double‐strand break repair, and sensitizes PanNET models to radiation, supporting SIRT7 as a ...

Singapore signalling: the 2012 hedgehog pathway cocktail [PDF]
The ‘Hedgehog Signalling in Development Evolution and Disease’ conference took place in Biopolis, Singapore, in March 2012. Organized by Phil Ingham with help from Xinhua Lin, Mary‐Ann Price and Fred de Sauvage, it brought leading researchers together to discuss the latest findings and exchange ideas on every aspect of hedgehog signalling.

Hedgehog Signaling and Truncated GLI1 in Cancer
The hedgehog (HH) signaling pathway regulates normal cell growth and differentiation. As a consequence of improper control, aberrant HH signaling results in tumorigenesis and supports aggressive phenotypes of human cancers, such as neoplastic ...

The Hedgehog Signaling Pathway is Expressed in the Adult Mouse Hypothalamus and Modulated by Fasting
The hedgehog signaling pathway is best known for its role in developmental patterning of the neural tube and limb bud. More recently, hedgehog signaling has been recognized for its roles in growth of adult tissues and maintenance of progenitor cell ...

Boosting Sensory Nerve‐to‐Bone Interactions Enhances Hedgehog Mediated Calvarial Bone Repair
Boosting sensory nerve activity via TrkA agonism strongly accelerates calvarial bone repair in adult mice. Furthermore, single‐cell RNA sequencing and neuron–bone interactome analyses identify these sensory neurons as a direct neural source of Hedgehog pathway ligands. Consequently, these ligands drive osteoblast differentiation of skeletal progenitors,

The role of Sulf1 during Sonic hedgehog mediated neural patterning [PDF]
Heparan sulphate proteoglycans (HSPGs) are large molecules distributed ubiquitously, both at the cell surface and within the extracellular matrix. These molecules are known to play essential roles in developmental cell signalling, and the differential ...

Single‐cell RNA sequencing verified the presence of the VIM+ biliary epithelial cell (BEC) in the bile ducts of NAS patients. The hypoxia/TGF‐β‐CREM‐VIM axis mediated phenotypic switch toward VIM+ BEC is validated using a hypoxia/TGF‐β‐stimulated cellular model, a rat liver transplantation model, BEC‐specific Crem conditional knockout rats, and BEC ...

Shifted from Wnt to Hedgehog Signaling Pathways [PDF]
Two papers in recent issue of Developmental Cell (Glise et al. 2005; Gorfinkiel et al. 2005) have shown that Shifted, a Drosophila ortholog of Wnt Inhibitory Factor (WIF), modulates the distribution of Hedgehog protein in the wing imaginal disc through a Wnt-independent mechanism.
