Two-step laser activation of hematoporphyrin derivative

Abstract A new photoactivation mechanism of hematoporphyrin derivative is presented. This process, based on the sequential absorption of two photons, leads to the production of cytotoxic radicals of HpD. The cytocidal efficiency is demonstrated by in vitro experiments.

Abstract Absorption and fluorescence properties of hematoporphyrin derivative in different solvent systems are investigated. The presence of a large amount of stable aggregates is demonstrated. The existence of these aggregates represents one major difference between hematoporphyrin derivative and hematoporphyrin free base.

HPD-A, the putative active biologic component of HPD, photosensitized lysis of large phosphatidylcholine liposomes and lipid peroxidation in small liposomes with oxygen enhancement ratios from 4 to 5. Unambiguous proof was obtained that this damage was mediated by singlet oxygen.

A dihematoporphyrin ether-free hematoporphyrin derivative has been prepared by a base-catalysed dehydration of hematoporphyrin with sodium hydroxide. The identification was performed by HPLC and mass spectroscopy (FD-MS). The reaction of hematoporphyrin with 1 M sodium hydroxide for 24 h yields more than 90% of the monomeric porphyrins.
