Genome Wide Association Studies (GWAS) has identified HNF1B located at chromosome 17q12 as foremost risk gene for prostate cancer susceptibility in multi-ethnic populations. This thesis characterises the HNF1B transcript variants along with defining their expression pattern and functional attributes in prostate cancer.

Heterozygous mutations in the transcription factors hepatocyte nuclear factor (HNF)-1α and -1β result in MODY (maturity-onset diabetes of the young). Despite structural similarity between HNF-1α and -1β, HNF-1β mutation carriers have hyperinsulinemia, whereas HNF-1α mutation carriers have normal or reduced insulin concentrations.

The MRP4 transporter exports several drugs and signaling molecules. Here, we identified key promoter elements regulating basal MRP4 expression. Using reporter assays, we defined a conserved region with essential Sp1 and contributory Ets sites, which controlled basal MRP4 expression.
