SEPARATION OF THE CYCLOPEPTIDE HETEROPHYLLIN B BY HIGH-SPEED COUNTERCURRENT CHROMATOGRAPHY AND ITS APPLICATION AS A NEW STATIONARY PHASE FOR CAPILLARY GAS CHROMATOGRAPHY

Analytical Letters, 2002
From the root of Pseudostellaria heterophylla a cyclopeptide, heterophyllin B, was isolated by high-speed countercurrent chromatography. Fused-silica capillary columns containing heterophyllin B as a main stationary phase component were prepared. The chromatographic selectivity of heterophyllin B showed toward aromatics.

Anti-neoplastic effect of heterophyllin B on ovarian cancer via the regulation of NRF2/HO-1 in vitro and in vivo

Tissue and Cell
Heterophyllin B (HB) is a cyclic peptide with anti-neoplastic effect on many cancers. However, its effect and mechanism of action in ovarian cancer cells are still unknown. The primary objective of this study was to assess the impact of HB on the proliferation of ovarian cancer (OC) cells and delve into the underlying mechanisms involved. We performed ...

Heterophyllin B alleviates cognitive disorders in APP/PS1 model mice via the spleen-gut microbiota-brain axis

International Immunopharmacology
Accumulating evidence implicates both the brain-spleen axis and the gut microbiota-brain axis in Alzheimer's disease (AD) pathogenesis. While our previous work demonstrated heterophyllin B (HB) rectifies splenic Th1/Th2 imbalance and ameliorates cognitive deficits in Aβ1-42-induced AD mice, its potential modulation of the vagus nerve-spleen circuit ...

Enzymatic cyclization of linear peptide to plant cyclopeptide heterophyllin B

Science in China Series B, 2006
The crude enzyme (PH-1) isolated from Pseudostellaria heterophylla by our group has catalyzed enzymatic cyclization of linear peptide NH2-Gly1-Gly2-Leu-Pro-Pro-Pro-Ile-Phe-COOH (4) into cyclopeptide heterophyllin B (HB) from plant for the first time.

Heterophyllin B, a cyclopeptide from Pseudostellaria heterophylla, enhances cognitive function via neurite outgrowth and synaptic plasticity

Phytotherapy Research, 2021
Neurite outgrowth‐induced construction of neural circuits and networks is responsible for memory generalization, consolidation, and retrieval. In this study, we found that the traditional Chinese medicine Pseudostellaria heterophylla promoted neurite regrowth and enhanced cognitive function in normal mice.

An in vitro system to study cyclopeptide heterophyllin B biosynthesis in the medicinal plant Pseudostellaria heterophylla

Plant Cell, Tissue and Organ Culture (PCTOC), 2011
Plant cyclopeptides are a large group of small molecule metabolites found in a wide variety of plants, including traditional Chinese medicinal plants. Many of the cyclopeptides have highly unusual structures and potent biological activities. However, the majority of the cyclopeptides have not been studied for their biosynthetic mechanisms.

LC‐ESI‐MS/MS analysis and pharmacokinetics of heterophyllin B, a cyclic octapeptide from Pseudostellaria heterophylla in rat plasma

Biomedical Chromatography, 2015
AbstractHeterophyllin B (HB) is a cyclic octapeptide isolated from Pseudostellaria heterophylla. HB is used as the quality control index for evaluating P. heterophylla in the Chinese Pharmacopoeia. A rapid and sensitive LC‐ESI‐MS/MS method was developed and validated for the analysis of HB in rat plasma.

PDL1 Monoclonal Antibody-Modified mPEG–PLGA-NH2 Nanocomposite Loaded with Heterophyllin B for the Treatment of Gastric Cancer

Journal of Medicinal Chemistry
Immune checkpoint inhibitors (ICIs) show effectiveness in gastric cancer (GC) treatment, yet enhancing their efficacy remains challenging. We encapsulated the anticancer compound Heterophyllin B (HB) in mPEG-PLGA-NH2 nanoparticles (NPs), modified with αPDL1, forming mPEG-PLGA-NH2@HB-αPDL1 NPs.
