Proteostasis collapse is a driver of cell aging and death. [PDF]
What molecular processes drive cell aging and death? Here, we model how proteostasis-i.e., the folding, chaperoning, and maintenance of protein function-collapses with age from slowed translation and cumulative oxidative damage.

Streamlined One‐Pot Synthesis of Fused Pyrroles: A Three‐Component Approach
New modified pyrrole derivatives, namely tetrahydroindol‐4‐ones and pyrrolo[2,3‐d]pyrimidines, functionalized at position 3, were synthesized through a one‐pot multicomponent condensation. Stabilized dimedone enaminones derived from α‐amino acids and their esters (or 6‐amino‐1,3‐dimethyluracil), arylglyoxals, and methylene‐active compounds ...

ApoJ regulates endothelial lipase activity and stability
Abstract Endothelial lipase (EL) is a key regulator of high‐density lipoprotein (HDL) metabolism. Many aspects of EL function remain incompletely understood due to challenges in purifying active EL. This study identifies apolipoprotein J (ApoJ) as a novel chaperone for EL, crucial for its solubility and activity.

From the cytosol to the inner membrane: biogenesis of the mitochondrial carrier family
Abstract Mitochondrial carrier proteins are essential for cellular physiology as they are active in a wide range of metabolic pathways including production of cellular energy, amino acid synthesis, redox balance and ion homeostasis. The double membrane of mitochondria provides a tightly gated environment through which carrier proteins facilitate the ...

This work reviewed the direct and indirect effects of fires on microbial communities and the resilience strategies microbes use to withstand high temperatures . ABSTRACT Fire, whether occurring on the surface or underground, significantly influences soil microbial dynamics by reshaping community composition, functional diversity and overall soil and ...

Single‐cell transcriptomics reveals that the human retina expresses a broad repertoire of steroid hormone biosynthesis and estrogen signalling genes. Functional validation in retinal explants shows that local estrogen synthesis is neuroprotective, preventing photoreceptor death and limiting microglial activation. Abstract Steroid hormones, particularly

Abstract The purpose of this exploratory investigation was to determine if acute post‐exercise skeletal muscle myosin heavy chain fragmentation (MyHCfrag) coincides with alterations in molecular chaperones and proteolytic enzymes, select markers of mammalian target of rapamycin complex 1 (mTORC1) signalling, and/or specific gene expression signatures ...

Heat shock induces Arc/Arg3.1 Cys34‐Cys159 disulfide crosslinking, promoting oligomerization and Hsp70‐CHIP‐dependent ubiquitination followed by proteasomal degradation during recovery. Disrupting disulfide formation (C159A) impairs CHIP‐mediated ubiquitination and stabilizes Arc/Arg3.1, whereas HSF1 loss limits inducible Hsp70 and leads to Arc ...
