Recent advances in the construction of humanized animal models and applications in translational medicine

open access: yesAnimal Models and Experimental Medicine, EarlyView.
Substantial advances have been achieved in the development of humanized mouse models, which have proven highly valuable in evaluating cancer immunotherapies and elucidating the mechanisms of infectious diseases. There is now a growing shift in research toward larger animal models—such as pigs—that offer greater physiological similarity to humans ...

Animal models in molecular biology research: Challenges, ethical imperatives, and the path to human‐relevant translation

open access: yesAnimal Models and Experimental Medicine, EarlyView.
The graphical abstract shows how molecular biology research has shifted from using traditional animal models toward using methods that are more relevant to humans. It points out the main problems, differences between species, difficulty in reproducing results, moral issues, and lack of infrastructure that make translational accuracy harder to achieve ...

Humanized Mouse Models of HIV Infection

open access: yes, 2011
Because of the limited tropism of HIV, in vivo modeling of this virus has been almost exclusively limited to other lentiviruses such as SIV that reproduce many important characteristics of HIV infection.

Nonviral Gold Nanoparticle‐Mediated Delivery of CRISPR‐Cas9 Ribonucleoprotein and Long DNA Transgenes Into Primary Blood Cells

open access: yesAdvanced NanoBiomed Research, EarlyView.
CRISPR/Cas9 has revolutionized the field of gene therapy, but delivery remains an outstanding issue. We propose a nonviral gold‐nanoparticle platform for co‐delivery of CRISPR/Cas9 ribonucleoprotein and long 2.1 kilobase dsDNA transgene constructs. This CRISPR‐AuNP is inexpensive to produce and mediate gene editing and DNA delivery in T cells and CD34+

Serological Profiles of Systemic Lupus Erythematosus in Humanized Mice and Pristane-Induced Lupus Models

open access: yesFolia Medica Indonesiana
Highlights: 1. This study compared the serological markers of pristane-induced mice to humanized mouse models of lupus achieved by transplanting stem cells from lupus patients, which is a novel method in Indonesia. 2.

Comparison of Receptor‐Mediated Endocytosis and Its Application to Enhance DNA Transfection by TFAMoplex

open access: yesAdvanced NanoBiomed Research, EarlyView.
We developed an assay to distinguish cellular binding from internalization. Compatible with microscopy and high‐throughput screening, the method identifies ligand‐mediated uptake. Applying top candidates to a protein‐based DNA carrier enhanced transfection efficiency, providing a rational strategy to improve non‐viral gene delivery systems.

Adenine base editing-mediated exon skipping restores dystrophin in humanized Duchenne mouse model

open access: yesNature Communications
Duchenne muscular dystrophy (DMD) affecting 1 in 3500–5000 live male newborns is the frequently fatal genetic disease resulted from various mutations in DMD gene encoding dystrophin protein. About 70% of DMD-causing mutations are exon deletion leading to

Establishment of Humanized EFNB2/B3 Murine Models for Investigating Nipah Virus Pathogenesis

open access: yesAnimal Research and One Health, EarlyView.
ABSTRACT Nipah virus (NiV) is a zoonotic paramyxovirus classified as a WHO priority pathogen, which causes severe respiratory disease and encephalitis in humans with high mortality. Currently, no licensed vaccines or specific antiviral therapies are available.

Establishment of a humanized mouse model: technical insight and applications

open access: yes
openHumanized mouse models have emerged as an important tool for cancer research. These models enable the grafting of human immune and cancer cells, allowing the study of the complex interaction between the immune system and cancer. The work presented in
