METTL3 Increases Temozolomide Resistance in MGMT‐Negative Glioblastoma by Enhancing CBX5 mRNA Stability in an m6A‐IGF2BP2‐Dependent Manner

METTL3‐dependent m6A modification and IGF2BP2‐associated CBX5 mRNA stabilization help maintain CBX5 expression in MGMT‐negative glioblastoma. During temozolomide (TMZ) treatment, CBX5 contributes to ATM–RAD51‐associated signaling, limits persistent DNA damage and apoptosis, and thereby supports cell survival and TMZ resistance.

Single‐cell and spatial transcriptomic profiling of 69 adult‐type diffuse gliomas revealed seven recurrent malignant cell states. GBMs were enriched for astrocyte‐like progenitor, hypoxic, and proliferative states, with higher T‐ and myeloid‐cell infiltration but transcriptional immune suppression.

TP53 Mutation Heterogeneity Refines Prognostic Stratification in IDH‐Wildtype Glioma

This study reveals that TP53 mutations in IDH‐wildtype gliomas are prognostically heterogeneous. A variant allele frequency (VAF) ≥10% identifies a high‑risk subgroup, and mutations located in the β‑strand region of the DNA‑binding domain confer the poorest outcomes.
