The FGF/FGFR System in the Biology and Therapeutic Landscape of Pediatric CNS Tumors

open access: yesMedicinal Research Reviews, EarlyView.
ABSTRACT Central nervous system (CNS) tumors are the most common solid malignancies in children, comprising a highly heterogeneous group of neoplasms defined by distinct molecular alterations and clinical behaviors. Advances in molecular genetics have underscored the relevance of specific signaling pathways in driving pediatric tumorigenesis, among ...

Isocitrate Dehydrogenase-Wildtype Glioma Adapts Toward Mutant Phenotypes and Enhanced Therapy Sensitivity Under D-2-Hydroxyglutarate Exposure

open access: yesBiomedicines
Background/Objectives: Isocitrate dehydrogenase (IDH) mutations are hallmark features in subsets of gliomas, producing the oncometabolite D-2-hydroxyglutarate (2HG).

Interlaboratory performance testing on EPIC v2.0 CNS tumor profiling demonstrates high reproducibility of tumor classification but reveals the need for harmonized copy number variation reporting

open access: yesBrain Pathology, EarlyView.
Using the Infinium MethylationEPIC v2.0 array and Heidelberg Brain Tumor Classifier v12.8, 24 international laboratories achieved highly reproducible CNS tumor classification (97.9% correct; median β‐correlation r = 0.99), while copy number variation interpretation showed substantial interlaboratory variability, highlighting the need for harmonized CNV

Targeting distinct amino acid metabolic vulnerabilities in IDH-mutant and IDH-wildtype gliomas

open access: yesActa Neuropathologica Communications
Lower grade gliomas frequently harbor mutations in isocitrate dehydrogenase (IDH), which define biologically distinct tumor subtypes. Although IDH-mutant and IDH-wildtype gliomas share similar histological morphology, they display markedly different ...

ALT phenotype assessment in IDH‐mutant astrocytoma supports biologic consequence of ATRX missense mutations

open access: yesBrain Pathology, EarlyView.
A meaningful subset of IDH‐mutant astrocytoma harbors ATRX missense variants that retain ATRX IHC immunoreactivity yet exhibit functional ALT activation, supporting the biologic relevance of these missense mutations.

Clinical evaluation of the CrossNN DNA methylation classifier for central nervous system tumors

open access: yesBrain Pathology, EarlyView.
CrossNN demonstrated non‐inferiority to the Heidelberg CNS Tumor Methylation Classifier. A dual‐classifier approach increased the number of clinically informative classifications with correct tumor type assignment while maintaining a low rate of incorrect predictions.

Molecular Characteristics and Novel Progress in Precision Targeted Therapy of IDH-Mutant Gliomas

open access: yes罕见病研究
Mutation of the isocitrate dehydrogenase (IDH) gene is a critical driver event in diffuse gliomas. By catalyzing the production of an abnormal metabolite, 2-hydroxyglutarate (2-HG), IDH mutation triggers a series of molecular alterations, including ...

Complement receptor C3aR marks heterogeneous tumor‐associated macrophage states associated with improved survival in IDH‐wildtype glioblastoma

open access: yesBrain Pathology, EarlyView.
C3aR expression in IDH‐wildtype glioblastoma is predominantly associated with the myeloid/macrophage compartment and heterogeneous macrophage states. C3aR‐high tumors are enriched among MGMT promoter‐methylated tumors and show improved overall survival, although the prognostic association requires independent validation.

ISG20 promotes local tumor immunity and contributes to poor survival in human glioma

open access: yesOncoImmunology, 2019
Recent evidence has confirmed that a mutation of the isocitrate dehydrogenase (IDH) gene occurs early in gliomagenesis and contributes to suppressed immunity.
