Human and Murine IL2 Receptors Differentially Respond to the Human-IL2 Component of Immunocytokines
OncoImmunology, 2019 The humanized immunocytokine, hu14.18-IL2 (ICp), leads to the immune cell-mediated destruction of GD2-expressing tumors in mouse models, resulting in potent antitumor effects with negligible IL2-related toxicity. In contrast, when ICp is used clinically,

Stimulating the Antitumor Immune Response Using Immunocytokines: A Preclinical and Clinical Overview
PharmaceuticsCytokines are immune modulators which can enhance the immune response and have been proven to be an effective class of immunotherapy. Nevertheless, the clinical use of cytokines in cancer treatment has faced several challenges associated with poor ...

Immunocytokines in Cancer Immunotherapy: Opportunities and Challenges
Frontiers in OncologyThe clinical use of cytokines is restricted by dose-limiting toxicities (DLT), short half-life, and rapid renal clearance, which collectively hinder administration at therapeutically effective doses.

Utilizing Immunocytokines for Cancer Therapy
Antibodies, 2021 Cytokine therapy for cancer has indicated efficacy in certain diseases but is generally accompanied by severe toxicity. The field of antibody–cytokine fusion proteins (immunocytokines) arose to target these effector molecules to the tumor environment in ...

Design and Characterization of Novel Antibody-Cytokine Fusion Proteins Based on Interleukin-21
Antibodies, 2022 Interleukin-21 (IL21) is a pleiotropic cytokine involved in the modulation of both innate and adaptive immunity. IL21 is mainly secreted by natural killer (NK) and activated CD4+ T-cells.

Targeted Cytokine Delivery for Cancer Treatment: Engineering and Biological Effects
Pharmaceutics, 2023 Anti-tumor properties of several cytokines have already been investigated in multiple experiments and clinical trials. However, those studies evidenced substantial toxicities, even at low cytokine doses, and the lack of tumor specificity.

Targeted Therapy With Immunoconjugates for Multiple Myeloma
Frontiers in Immunology, 2020 The introduction of proteasome inhibitors (PI) and immunomodulatory drugs (IMiD) has markedly increased the survival of multiple myeloma (MM) patients. Also, the unconjugated monoclonal antibodies (mAb) daratumumab (anti-CD38) and elotuzumab (anti-SLAMF7)

Improved intratumoral penetration of IL12 immunocytokine enhances the antitumor efficacy
Frontiers in Immunology, 2022 Tumor-targeting antibody (Ab)-fused cytokines, referred to as immunocytokines, are designed to increase antitumor efficacy and reduce toxicity through the tumor-directed delivery of cytokines.

Frontiers in Immunology, 2018 Antibody-cytokine fusion proteins (immunocytokine) exert a potent anti-cancer effect; indeed, they target the immunosuppressive tumor microenvironment (TME) due to a specific anti-tumor antibody linked to immune activating cytokines.
