AbstractThis unit describes procedures for fragmentation of IgG to the monovalent Fab fragment using papain digestion, and to the bivalent F(ab′)2 fragment by pepsin digestion. Alternative methods of fragmentation to F(ab′)2 include use of papain that is preactivated with cysteine and use of the enzyme ficin.

Canadian Journal of Biochemistry, 1966Solutions of human γ-globulin undergo changes on storage resulting in the formation of fragments similar to those produced by the digestion of human γ-globulin with the enzymes papain and plasmin. Two fragments have been isolated from the naturally fragmented material which resemble the Fab and Fc fragments resulting from digestion by papain.

Employing immunoelectrophoresis with antisera to whole human serum and to Fab‐fragments of IgG, a marked precipitation line, likely to represent such fragments, was evident in the cathodal or γ‐region.

Coprecipitation of some cryomacroglobulins with immunoglobulins and their fragments
Archives of Biochemistry and Biophysics, 1965Abstract Four cryomacroglobulins, isolated from different pathological sera, were found to coprecipitate on cooling with normal IGG. Coprecipitation occurred also with the immunoelectrophoretically fast component, and to a much lesser extent with the slow component, of papain digests of IGG and the cryomacroglobulins.

Immunoglobulin genes in DNA restriction fragments
Biochemistry, 1978We have investigated the organization of immunoglobulin genes in mice. High molecular weight DNA from myelomas and Krebs ascites cells was cleaved with EcoRI restriction endonuclease and fractionated using preparative agarose gel electrophoresis. Each fraction was then hybridized to an immunoglobulin mRNA or a cDNA transcribed from the mRNA.
