Tumor‐Derived LAMB3 Drives Immunosuppressive LRRC15+ Fibroblast Formation During Pancreatic Ductal Adenocarcinoma Development

A single‐cell atlas of pancreatic ductal adenocarcinoma development reveals progressive ductal‐fibroblast‐immune crosstalk. Tumor‐derived LAMB3 drives the formation of immunosuppressive LRRC15+ fibroblasts through the ITGB1/FAK/MAPK/FOSL2 signaling. Glycolytic reprogramming upregulates LAMB3 and correlates with LRRC15+ fibroblast enrichment.

Enhancing CAR‐T Cell Efficacy in Solid Tumors by Inhibiting CCL5/VEGF‐Mediated Angiogenesis

This study reveals that CAR‐T cells in solid tumors produce CCL5, which paradoxically induces VEGF and angiogenesis to promote tumor growth. RBM10 deficiency promotes anti‐PD‐1 resistance in lung adenocarcinoma by altering STING alternative splicing, which enhances CCL7 secretion and CCR2‐dependent M2 macrophage polarization. A positive feedback loop via mitochondrial transfer sustains this immunosuppression.

Aptamer‐Engineered Liposomal Platform Enables in Situ cDC1 Vaccination to Potentiate Immunotherapy in Prostate Cancer

Prostate cancer is immunologically ‘cold’, with scarce, dysfunctional type 1 conventional dendritic cells (cDC1s) that limit T cell priming. We introduce an aptamer‐targeted liposomedelivering FMS‐like tyrosine kinase 3 ligand (Flt3L) and chlorin e6 (Ce6). Ultrasound induces antigen release and cDC1s recruitment, creating an in situ cDC1 vaccine.
