ABSTRACT Osteoarthritis (OA) is a common degenerative joint disease characterized by chondrocyte dysfunction. In this study, we explored the function and mechanism of ubiquitin‐specific protease 7 (USP7) in chondrocyte ferroptosis in OA. The USP7, TRPV1, collagen II, and GPX4 levels in knee joint tissue were detected using immunohistochemistry ...

A Network‐Based Approach to Understanding Key Signaling Pathways in Intervertebral Disc Biology
A literature‐based signaling network capturing the most relevant pathways in human nucleus pulposus cells was first constructed from curated biological evidence. The network was then translated into a system of ordinary differential equations to derive steady‐state solutions and define median baseline activity levels.

Kontrolle der monozytären Interleukin-1beta-Freisetzung durch alpha1-Antitrypsin
The release of the pro-inflammatory cytokine interleukin-1beta (IL-1beta) by monocytes can contribute to the development of a systemic inflammation and therefore requires strict control. In addition to its function as an anti-protease, the acute phase protein alpha1-antitrypsin (AAT) has multiple anti- inflammatory properties that can be potentially be

Abstract Purpose This study aims to analyze the effect of pro-inflammatory cytokine-stimulated human annulus fibrosus cells (hAFCs) on the sensitization of dorsal root ganglion (DRG) cells. We further hypothesized that celecoxib (cxb) could inhibit hAFCs-induced DRG sensitization. Methods

Inhibition of the NLRP3 Inflammasome With MCC950 Improves Gut Health in Huntington's Disease Mice
The NLRP3 inflammasome in HD (Huntington's disease) peripheral pathology (in gastrointestinal complications) is vastly unexplored. In this preclinical study, non‐invasive readouts of gut function assessment revealed that inhibiting NLRP3 inflammasome by chronic administration of MCC950 by oral route softened the faecal pellets, improved faecal output ...

Cytokine responses to Dengue Infection among Puerto Rican Patients
Recently, a strong correlation between high concentration of tumor necrosis factor (TNFalpha) in blood and severity of dengue hemorrhagic fever/dengue shock syndrome has been reported from Asia and the Pacific.

This study identifies platelet‐derived extracellular vesicles as key immunometabolic regulators in COVID‐19. The delivery of tRF‐His‐GTG‐1 to neutrophils activates TLR8mTOR signaling, disrupts lipophagy, and amplifies NET‐mediated inflammation. Importantly, targeting this axis restores neutrophil homeostasis, offering a potential therapeutic strategy ...

Radiotherapy induces tumor cells to release microparticles (RT‐MPs) into the circulation. The mitochondrial DNA carried by these RT‐MPs activates the STING/NLRP3/GSDMD axis in splenic neutrophils, triggering IL‐1β secretion. This, in turn, enhances dendritic cell function and facilitates the formation of cytotoxic T lymphocytes, thereby promoting ...

Obesity and Natural Products: Mechanisms, Therapeutic Potential, and Future Directions
Natural products such as EGCG, berberine, curcumin, and capsaicin modulate key metabolic pathways including AMPK activation, adipogenesis inhibition, and inflammatory signaling. While clinical trials demonstrate modest benefits on body weight and metabolic parameters, translational challenges related to bioavailability, dose feasibility, and regulatory
